Aim: Determine an optimal set of the most effective methods of identification and intraspecies typing ofcausative agents ofglanders and melioidosis. Materials andmethods. Bacteriologic, immunochemical, molecular-genetic methods were used.

Results: A possibility to identify collection strains of pathogenic and closely related Burkholderia in semiautomatic systems is studied. Means of detection of informative variable genome segments ofthe specified microorganisms were developed, methods of their genetic typing were selected. Effectiveness of application of precipitating mAbs for differentiation of Burkholderia was established. Data on diagnostic possibilities of immunoglobulins fluorescing based on monoclonal antibodies of various etiotropic directionality for detection and identification of B. mallei and B. pseudomallei are generalized. Experimental series of amplification test-systems for identification of glanders and melioidosis causative agents in real-time PCR format are created.

Conclusion: A number of methods for identification and typing of glanders and melioidosis causative agents is proposed.

Isolation and identification of the causal agent of gummy stem blight disease in Cucumis sativus caused by a bacterial pathogen in China.

Recently, a new bacterial disease was detected on cucumber stalks. In order to study the pathogenesis of this disease, the pathogenic bacteria were isolated and identified on the basis of morphological and molecular characteristics, and further analyzed for pathogenicity and antagonistic evaluation. Pathogenicity analysis showed that HlJ-3 caused melting decay and cracking in cucumber stems, and the strain reisolated from re-infected cucumber stalks was morphologically identical to HlJ-3 colonies, which is consistent with the Koch's postulates.

To investigate the effect of the filter device on the cleanliness of molten steel and the flow field distribution within the tundish. The tundish filters were designed into five groups of pores with 20°, 25°, 30°, 35° and 40° elevation angles, and the flow field distribution and impurity removal rate of molten steel were calculated by Discrete Phase Model (DPM). The results showed that the removal rate of impurity in the molten steel could be significantly improved by using the tundish filter with elevation angle.
