Unlabelled: The phosphatidylinositol 3-kinases (PIK3s) are lipid kinases. Mutation in the exon 9 and exon 20 determined as a predictive factor in anti-HER-2 therapy. In some countries, such as Singapore, China, and Peru, PIK3CA exon 9 E545A was reported to produce the highest rate of mutation. In this research, we developed and optimized PIK3CA exon 9 E545A detection methods with intercalating dye SYBR Green I based on the Tm Shift approach by using prepared recombinant plasmid pGEMT-easy PIK3CA exon 9 and PIK3CA exon 9 E545A. Recombinant plasmid was used due to the limited number of samples.

Methods: Recombinant plasmid was prepared based on manufactured procedures, and this process was then followed by Tm prediction with Poland software, Tm Shift SYBR Green I development, and its characterization (reproducibility, repeatability, sensitivity, qPCR efficiency, and qPCR amplification), respectively.

Result: A method for PIK3CA E545A detection based on TM shift SYBR Green I has been successfully developed. The melting temperature for PIK3CA exon 9 was 78.1 ± 0.1 °C, while that for PIK3CA exon E545A was 80.20 °C. The Tm of mutant was the same as that predicted using Polland Software. The reproducibility of the methods was high, with the coefficient values for inter and intra assays were below 10% with a high sensitivity at 1%, while R 0.99 and PCR efficiency was 97.75%.

Conclusion: The results presented here demonstrate that the PIK3CA exon 9 E545A detection method has a good sensitivity and efficacy assay, which proves that the method has a high diagnostic accuracy in breast cancer.

Background: We aimed to investigate the clinical and molecular characteristics of different degrees of human epidermal growth factor receptor 2 (HER2) protein expression in HER2-negative breast cancer and the related factors affecting the efficacy of neoadjuvant chemotherapy in HER2-low breast cancer patients.

Methods: The study endpoint was pathological complete remission (PCR). Blood specimens and fresh cancer tissue samples were collected before neoadjuvant chemotherapy for whole-exon sequencing (WES) and RNA sequencing (RNA-seq), and patients were divided into a human epidermal growth factor receptor 2 (HER2)-low group and a HER2-0 group according to their HER2 expression status via bioinformatics analysis.

  • Lung cancer is the leading cancer globally, with EGFR mutations common in female NSCLC patients, necessitating targeted therapies like TKIs, though resistance often develops.
  • A case study details a 63-year-old woman with both lung adenocarcinoma and renal cancer; she initially responded well to standard treatments but faced resistance.
  • The use of furmonertinib, a third-generation EGFR-TKI, resulted in a remarkable 27 months of progression-free survival, showcasing its effectiveness against complex resistance pathways.

Objective: To explore the gene mutations of Langerhans cell histiocytosis in children, and to analyze the correlation of mutation with clinical features and prognosis of LCH, so as to provide reference for clinical diagnosis and treatment.

Methods: Fluorescence PCR was used to detect gene mutations in paraffin-embedded tissue samples from 78 children with LCH, and the correlation of mutation with clinical characteristics and prognosis of LCH in children was analyzed.

Results: Among the 78 children, 41 cases (52.

Background/aim: Soft tissue sarcoma (STS) is a mesenchymal tumor affecting multiple organs in dogs. Previous studies identified activation of the phosphatidylinositol-3 kinase (PI3K)/protein kinase B (PKB, AKT) pathway in canine STS cell lines and clinical samples, but the underlying mechanism remains unclear. This study investigated PTEN loss, PIK3CA mutation, and EGFR over-expression as potential drivers of PI3K/AKT pathway activation in STS.

Article Synopsis
  • The study compares the effectiveness of ultrasensitive real-time PCR and droplet digital PCR (ddPCR) in detecting specific mutations associated with breast cancer in primary tumors and liquid biopsy samples.
  • The research involved analyzing genetic material from 42 tumor samples and 29 plasma samples from patients with ER+ metastatic breast cancer, as well as samples from healthy donors.
  • Results showed that both methods provided similar detection rates for certain mutations in tumor samples, with ultrasensitive real-time PCR performing better in plasma-cfDNA samples, indicating potential for non-invasive testing in cancer management.
