A new isoflavone, milletenol A (), along with four known flavonoids (-) were isolated from the seeds of . The structure of was established by extensive spectroscopic methods while known compounds were identified by comparisons with literature data. Compound and showed significant anti-inflammatory activities against nitric oxide production in LPS-induced RAW264.7 macrophages. The state of CuSO-stimulated inflammation was effectively alleviated by compound in zebrafish. However, no significant cytotoxicity against human breast cancer cells was observed among all isolates.

Total Synthesis and Anti-Inflammatory Activity of Tectoridin and Related Isoflavone Glucosides.

J Nat Prod

December 2024

Anti-infective Agent Creation Engineering Research Centre of Sichuan Province, School of Pharmacy, Chengdu University, Chengdu 610106, China.

The first total syntheses of four isoflavone glucosides, tectoridin (), tectoridin A (), tectorigenin 7--β-d-glucopyranosyl-12--β-d-glucopyranoside (), and isotectroigenin 7--β-d-glucopyranoside (), have been accomplished. Key steps in our synthetic approach include a regioselective halogenation reaction, followed by methanolysis to introduce the -OCH group into isoflavone frameworks and a PTC-promoted stereoselective glycosidation to establish glycosidic bonds. The synthesized isoflavone glucosides (-) and their corresponding aglycones ( and ) were evaluated for anti-inflammatory activity against nitric oxide (NO), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and interleukin-1 β (IL-1β) in lipopolysaccharide (LPS)-induced RAW264.

Background: Among the bioactive isoflavones identified from different plants is formononetin. Formononetin's antioxidant, anti-inflammatory, and anti-cancer qualities have all received a lot of attention lately. The goal of the current investigation was to examine formononetin's antifungal and antibacterial activity against Candida albicans and Enterococcus faecalis in vitro, respectively.

The investigation of the methanol extract of the flowers of Jacaranda mimosifolia led to the isolation and characterisation of fourteen secondary metabolites (1-14) by the means of chromatographic and spectroscopic analysis. These compounds include eight flavonoids sorted as two flavonols (1-2), three flavones (3, 7-8), and three isoflavones (4, 9-10), two glucoiridoids (5-6), and four triterpenoids sorted as one oleanane (11), one hopane (12), and two lupane (13-14). The in vitro antioxidant potential of all the isolated compounds was evaluated using the ABTS- and FRAP- methods, which demonstrated concentration-dependent activity.

Roles of 4'--Methylalpinum Isoflavone on Activation of Microglia Induced by Oxysterols.

Int J Mol Sci

November 2024

Department of Pharmacology, School of Medicine, Pusan National University, Yangsan 50612, Gyeongnam, Republic of Korea.

Microglia play a crucial role as immune cells responsible for the brain's defense mechanisms. Similar to the actions of macrophages in the body, microglial cells elicit an inflammatory immune response in the brain. Recent papers highlight activated microglial cells as pivotal contributors to inflammatory responses in the brain, leading to damage to nerve tissue and the onset of Alzheimer's disease (AD).

Isoflavones are found in numerous plant species within the Leguminosae family; however, soy isoflavones are particularly significant in practice and have been extensively studied in recent years. The health-promoting potential of orally administered soy isoflavones is widely documented in the scientific literature, and many review articles have been developed to highlight their significance. However, it should be noted that soy-isoflavone-rich extracts and isolated soy isoflavones, such as genistein and daidzein, are also often applied topically as ingredients in many formulations, including face creams, tonics, and emulsions.
