Ratiometric temperature sensing with fluorescent thermochromic switches.

The connection of fluorescent chromophores to switchable heterocycles translates into molecular probes with ratiometric response to temperature. The opening and closing of their heterocyclic component equilibrates two emissive species with resolved fluorescence. Their relative emission intensities change monotonically with temperature to enable the visualization of thermal distributions at the microscale.

Firefly luciferases have been extensively used for bioanalytical applications, including their use as bioluminescent reporters, biosensors, and for bioimaging biological and pathological processes. Due to their intrinsic pH- sensitivity, in recent years we have demonstrated that firefly luciferases can also be harnessed as color- tuning sensors of intracellular pH. However, it is known that mammalian cells require temperatures higher than 36 °C, which red-shift the bioluminescence spectra of most firefly luciferases, decreasing their activities and the resolution of ratiometric pH analysis.

Molybdenum disulfide quantum dots (MoS2 QDs) is a new type of graphite like nanomaterial, which exhibited well chemical stability, unique fluorescence characteristics, and excellent biocompatibility. The conventional hydrothermal synthesis of MoS2 generally requires a long-term reaction at high temperature and high pressure. Herein, we have developed a simple and fast MoS2 QDs synthesis scheme using microwave heating, and further modified the surface of MoS2 QDs using 3-aminophenylboronic acid.
