Obsessive-compulsive disorder (OCD) is a severe, chronic neuropsychiatric disorder with a strong genetic component. The SLC1A1 gene encoding the neuronal glutamate transporter EAAT3 has been proposed as a candidate gene for this disorder. Gene variants affecting SLC1A1 expression in human brain tissue have been associated with OCD. Several mouse models fully or partially lacking EAAT3 have shown no alterations in baseline anxiety-like or repetitive behaviors. We generated a transgenic mouse model (EAAT3) to achieve conditional, Cre-dependent EAAT3 overexpression and evaluated the overall impact of increased EAAT3 expression at behavioral and synaptic levels. Mice with EAAT3 overexpression driven by CaMKIIα-promoter (EAAT3/CMKII) displayed increased anxiety-like and repetitive behaviors that were both restored by chronic, but not acute, treatment with fluoxetine or clomipramine. EAAT3/CMKII mice also displayed greater spontaneous recovery of conditioned fear. Electrophysiological and biochemical analyses at corticostriatal synapses of EAAT3/CMKII mice revealed changes in NMDA receptor subunit composition and altered NMDA-dependent synaptic plasticity. By recapitulating relevant behavioral, neurophysiological, and psychopharmacological aspects, our results provide support for the glutamatergic hypothesis of OCD, particularly for the increased EAAT3 function, and provide a valuable animal model that may open novel therapeutic approaches to treat this devastating disorder.

The early post-hatching phase remains to be one of the most vulnerable phases in broiler production. Some essential oils have been reported to improve gut health and growth in broiler chickens when applied to post-hatching diets. However, in-feed applications are unable to prevent the health challenges observed immediately after hatching.

Tetrastigma hemsleyanum (TH) has attracted much attention for its heat clearing and detoxification effects, but whether it can become an effective feed supplement in chickens remains unclear. Herein, a total of 120 male Jinhua yellow chickens (two-mth-old) were randomly divided into into four groups (CON, TH-L, TH-M, and TH-H) for a 56-day feeding trial to explore its effects on growth performance and underlying mechanism. Results revealed that dietary TH notably increased the average daily growth (ADG), and decreased the average daily feed intake (ADFI) and feed conversion ratio (FCR) in TH-H group during 29-56 days.

Article Synopsis
  • Depression is a common mental illness linked to glutamate system dysfunction, with a specific focus on the neuronal glutamate transporter EAAT3 as a potential contributor to depressive-like behaviors.
  • Research involving mice showed that those with increased EAAT3 expression in the forebrain were more resilient to developing depressive-like behaviors after undergoing chronic mild stress.
  • Results indicated that EAAT3 overexpression not only protected these mice from depressive symptoms but also preserved their memory and normal synaptic activity, suggesting EAAT3's crucial role in mitigating the effects of chronic stress on mental health.

Article Synopsis
  • The study investigated the effects of a microencapsulated blend of botanicals and organic acids on digestive enzymes and nutrient transporters in broilers, revealing beneficial impacts on gut health and growth performance.
  • 288 birds were divided into a control group and a supplemented group, with significant improvements in feed efficiency and intestinal histology observed in the supplemented group after 21 days.
  • Key findings included enhanced activities of enzymes like maltase and sucrase and increased expression of nutrient transporters, indicating that the blend positively influences nutrient absorption in the broilers.

An experiment was conducted to assess the response of chicks to in-ovo injection of Bacillus subtilis (probiotic), raffinose (prebiotic), and their combinations. The study used 1,500 embryonated eggs allotted to 10 groups/ 6 replicates (150 eggs/group). The experimental treatments were: 1) un-injected control (NC); 2) sham (sterile distilled water) (PC); 3) probiotic 4 × 10CFU/egg (LBS); 4) probiotic 4 × 10CFU/egg (HBS); 5) prebiotic 2 mg/egg (LR); (6 prebiotic 3 mg/egg (HR); 7) probiotic 4 × 10CFU + prebiotic 2 mg/egg (LBS+LR); 8) probiotic 4 × 10CFU + prebiotic 3 mg/egg (LBS+HR); 9) probiotic 4 × 10CFU + prebiotic 2 mg/egg (HBS+LR); and 10) probiotic 4 × 10CFU + prebiotic 3 mg/egg (HBS+HR).
