The tissue engineering method of decellularization and recellularization has been successfully used in a variety of regenerative medicine applications. The protocols used to de/recellularize various organs and tissues are largely different. Here we describe a method to effectively engineer a bioartificial colon by completely removing original cells from human intestinal tissues followed by repopulating the acellular tissue matrix with cell cultures. This method provides a novel approach for human intestinal regeneration and can be used to identify potential cancer driver genes.

This work presents strong evidence supporting the use of decellularized human iliac arteries combined with adipose tissue-derived stem cells (hASCs) as a promising alternative for vascular tissue engineering, opening the path to future treatments for peripheral artery disease (PAD). PAD is a progressive condition with high rates of amputation and mortality due to ischemic damage and limited graft options. Traditional synthetic grafts often fail due to poor integration, while autologous grafts may be unsuitable for patients with compromised vascular health.

Chronic kidney disease (CKD) is a prevalent global health issue, primarily caused by glomerular dysfunction, diabetes, endovascular disorders, hypertensive nephrosclerosis, and other vascular diseases. Despite the increase in available organ sources, significant challenges remain in securing organ compatibility, prompting extensive research into creating a bio-artificial kidney free from immune rejection. In this study, a bio-engineered kidney was established using a stem cell chemoattractant within a bioreactor system; rBMSCs were used to recellularize the decellularized kidney scaffold coated with SDF-1α/AKI-CKD cytokine juice under mimic-hypoxic conditions as these chemokines and cytokines are crucial for the cell migration.

Introduction: Uterus bioengineering offers a potential treatment option for women with uterine factor infertility and for mitigating the risk of uterine rupture associated with women with defective uterine tissue. Decellularized uterine tissue scaffolds proved promising in further in vivo experiments in rodent and domestic species animal models. Variations in the extracellular matrix composition among different species and adaptations of the decellularization protocols make it difficult to compare the results between studies.

Background: End-stage renal disease is a growing global health issue, disproportionately impacting low- and middle-income countries. While kidney transplantation remains the best treatment for end-stage renal disease, access to this treatment modality is limited by chronic donor organ shortages. To address this critical need, we are developing transplantable bioengineered kidney grafts.

Decellularized extracellular matrix sheets (dECMSs) produced by stem cells have attracted attention because they preserve the natural biological activity of the ECM to direct lineage-specific differentiation with less immunogenicity. As a core ECM protein, laminin modulates cellular phenotype and differentiation. Nevertheless, no studies thus far have explored the distribution and abundance of laminins in diverse dECMSs.
