N-Linked glycopeptides have highly diverse structures in nature. Herein, we describe the first synthesis of rare multi-antennary N-glycan bearing glycan chains on 6-OH of both α1,6- and α1,3-linked mannose arms. To expedite divergent generation of N-glycan structures, four orthogonal protective groups were installed at the branching points on the core tetrasaccharide, which could be removed individually without affecting one another. In addition, the synthetic route is flexible, allowing a bisecting glucosamine moiety to be introduced at a late stage of the synthesis, further expanding the diversity of sequences that could be achieved. The bisecting glucosamine unit significantly reduced the glycosylation yields of adjacent mannoses, which was attributed to steric hindrance imposed by the glucosamine based on molecular modelling analysis. The N-glycans were then transformed to oxazoline donors and ligated with a glycopeptide acceptor from haptoglobin promoted by the wild type -β--acetylglucosaminidase (Endo-A). Endo-A exhibited interesting substrate preferences depending on donor sizes, which was rationalized through molecular dynamics studies. This is the first time that a glycopeptide bearing a bisecting -acetyl glucosamine (GlcNAc), the rare N-glycan branch, and two Lewis trisaccharide antennae was synthesized, enabling access to this class of complex glycopeptide structures.

Article Synopsis
  • Aggregations of misfolded proteins are linked to various diseases, including a rare condition called Autosomal dominant neurohypophyseal diabetes insipidus (ADNDI) caused by mutations in the AVP-NPII gene leading to cellular dysfunction due to accumulated proteins in the endoplasmic reticulum (ER).
  • The study aimed to investigate the formation of amyloid-like aggregates from mutant precursor proteins linked to specific mutations found in ADNDI patients, examining factors such as glycosylation patterns and degradation pathways.
  • Results indicated that mutant precursors maintained their glycan structure, were involved in degradation pathways, formed oligomers, and demonstrated fibril formation in bacterial models, providing insights into the mechanisms underlying ADNDI.

Article Synopsis
  • N-glycans show significant diversity in commercial products, prompting a need to reassess species designations and product authenticity.
  • An analysis of 172 products revealed that only a small percentage (9%) matched expected N-glycan patterns, with most exhibiting unique or rare structures.
  • The findings highlight a complex relationship between N-glycan diversity and product identity, encouraging further exploration of microalgal glycans in classification efforts without questioning the benefits or integrity of the products.

Article Synopsis
  • This study focuses on identifying plasma IgG N-glycan biomarkers, specifically fucosylation, to enhance risk assessment and survival prediction in patients with pulmonary arterial hypertension (PAH).
  • Researchers analyzed data from 622 PAH patients across two different cohorts and utilized mass spectrometry to profile IgG N-glycans, confirming the effectiveness of these biomarkers through various statistical models.
  • The findings indicate that IgG fucosylation is a strong and independent predictor of survival in PAH, and incorporating it into existing risk assessment models improves their predictive accuracy, especially for intermediate-risk patients.

Introduction: Although inguinal hernia (IH) is prevalent in elderly males, research on its specific diagnostic biomarkers is limited. Protein -glycosylation is one of the most important and ubiquitous post-translational modifications and often results in a remarkable heterogeneity of protein glycoforms. Protein -glycosylation often changes in a disease and holds great potential for discovering non-invasive biomarkers.

The treatment of defective glycosylation in clinical practice has been limited to patients with rare and severe phenotypes associated with congenital disorders of glycosylation (CDG). Carried by approximately 5% of the human population, the discovery of the highly pleiotropic, missense mutation in a manganese transporter ZIP8 has exposed under-appreciated roles for Mn homeostasis and aberrant Mn-dependent glycosyltransferases activity leading to defective N-glycosylation in complex human diseases. Here, we test the hypothesis that aberrant N-glycosylation contributes to disease pathogenesis of ZIP8 A391T-associated Crohn's disease.
