A series of 19 analogues of the antiproliferative naphthopyran LY290181 were prepared for structure-activity relationship studies. We found the best activities for test compounds bearing small substituents at the meta position of the phenyl ring. The mode of action of LY290181 and eight new analogues was studied in detail. The compounds were highly anti-proliferative with IC values in the sub-nanomolar to triple-digit nanomolar range. The new analogues led to G2/M arrest due to interruption of the microtubule dynamics. In 518A2 melanoma cells they caused a mitotic catastrophe which eventually led to apoptosis. The naphthopyrans also induced a disruption of the vasculature in the chorioallantoic membrane (CAM) of fertilized chicken eggs as well as in xenograft tumors in mice. In a preliminary therapy trial, the difluoro derivative 2b retarded the growth of resistant xenograft tumors in mice.

Berberine (BBR), an isoquinoline alkaloid abundant in Coptis chinensis, exhibits anti-tumor and hypoglycemic properties. The regulation of tumor cell homeostasis and metabolism is greatly influenced by Hypoxia-inducible factor-1α (HIF-1α). This research aims to elucidate whether BBR inhibits the progression of hepatocellular carcinoma (HCC) by modulating HIF-1α expression.

Atractylenolide I (ATL-I) can interfere with Colorectal cancer (CRC) cell proliferation by changing apoptosis, glucose metabolism and other behaviors, making it an effective drug for inhibiting CRC tumor growth. In this paper, we investigated the interactions between ATL-I and Keratin 7 (KRT7), a CRC-specific marker, to determine the potential pathways by which ATL-I inhibits CRC development. The KRT7 expression level in CRC was predicted online using the GEPIA website and then validated.

Glioblastoma (GBM) is a highly aggressive adult brain cancer, characterised by poor prognosis and a dismal five-year survival rate. Despite significant knowledge gains in tumour biology, meaningful advances in patient survival remain elusive. The field of neuro-oncology faces many disease obstacles, one being the paucity of faithful models to advance preclinical research and guide personalised medicine approaches.

Background: Oxaliplatin is the first-line chemotherapy for patients with colon cancer (CC). However, its resistance limits its therapeutic efficacy.

Methods: Oxaliplatin resistance-associated differentially expressed genes (DEGs) in the GSE42387 and GSE227315 datasets were identified through bioinformatics methods.

SEPT5 overexpression predicts poor prognosis and promotes progression through feedback regulation of HIF-1α in clear cell renal cell carcinoma.

Department of Biochemistry and Molecular Biology, School of Basic Medical Sciences, Kunming Medical University, Kunming, China. Electronic address:

Clear cell renal cell carcinoma (ccRCC), a predominant subtype of renal cell carcinoma, significantly contributes to the heightened morbidity and mortality in individuals diagnosed with urologic tumors. The challenges posed by high malignancy at the initial diagnosis of ccRCC, therapeutic resistance, and unfavorable patient prognosis remain largely unresolved. Our findings indicate that SEPT5 is upregulated in ccRCC and this upregulation is associated with an adverse prognosis for ccRCC patients.
