Objectives: Failed fertilization after intra-cytoplasmic sperm injection (ICSI) is mainly attributed to failed oocyte activation and can be overcome by artificial oocyte activation (AOA). The present study aims to compare outcomes of ICSI following two different assisted oocyte activation chemical procedures (SrCl and Ionomycin) in sibling oocytes of ICSI candidates.

Materials And Methods: From March 2015 until February 2016, 105 infertile men with 99-100% abnormal sperm morphology, irrespective of sperm motility, concentration, or origin (semen or testicular) were included in this study. Out of these, 66 couples accepted to be included in the study group (Ionomycin/ SrCl) and 39 couples requested routine AOA procedure (Ionomycin) as external control group. Primary outcomes of this study (fertilization, embryo quality, and post-implantation development) were compared between these groups.

Results: Significantly higher oocyte activation (67.90±3.6% vs. 51.16±3.6%, =0.004) and fertilization (65.23±3.63% vs. 49.65±3.63%, =0.008) rates were observed in sibling oocytes treated with Ionomycin in comparison to the SrCl sibling group. Percentage of top quality embryos was insignificantly higher in SrCl groups compared to the Ionomycin group (29.90±4.27 vs. 20.65±4.05%, =0.26).

Conclusion: Ionomycin may be superior to SrCl for inducing oocyte activation. However, SrCl may be a more efficient means to support the development of better quality embryos following ICSI.

Artificial oocyte activation (AOA) with Ca2+ ionophores is an experimental procedure that benefits patients who fail to obtain fertilized eggs. However, the impact of non-physiological Ca2+ increases on cellular events involved in egg-embryo transition and early development remains poorly understood. Using the mouse model, this study compares common Ca2+ ionophore protocols applied in clinical practice - one or two exposures to A23187 or a single exposure to ionomycin - focusing on embryonic development and cellular events associated with egg activation.

Oocyte quality is crucial for determining the subsequent embryo developmental capacity and reproductive outcomes. However, aging is detrimental to oocyte quality. Previous studies have demonstrated that soy isoflavones have positive effects on the reproductive performance of female pigs.

A limited number of female germ cells support reproduction in many mammals. The follicle, composed of oocytes and supporting granulosa cells, forms the basis of oogenesis. Crosstalk between oocytes and granulosa cells is essential for the formation, dormancy, re-awakening, and maturation of oocytes.

The mammalian target of rapamycin (mTOR) signaling pathway is activated by luteinizing hormone in preovulatory follicle. However, its impact on ovulation remains inadequately explored. Utilizing in vivo studies and in vitro fertilization, we demonstrated that the negative effect of inhibition of mTOR signaling by rapamycin on oocyte quality during the ovulatory phase, with a notable decrease in the total cell count of blastocysts, a reduction in gastrula size, and fetal degeneration on the 16th day of gestation while not affecting ovulated oocyte count or granulosa cell luteinization.

Female reproductive senescence results from the regulated depletion of a finite pool of oocytes called the ovarian reserve. This pool of oocytes is initially established during fetal development, but the oocytes that comprise it must remain quiescent for decades until they are activated during maturation in adulthood. In order for developmentally competent oocytes to populate the ovarian reserve they must successfully initiate both meiosis and oogenesis.
