In eukaryotic organisms, replication initiation follows a temporal program. Among the parameters that regulate this program in , chromatin structure has been at the center of attention without considering the contribution of transcription. Here, we revisit the replication initiation program in the light of widespread genomic noncoding transcription. We find that noncoding RNA transcription termination in the vicinity of autonomously replicating sequences (ARSs) shields replication initiation from transcriptional readthrough. Consistently, high natural nascent transcription correlates with low ARS efficiency and late replication timing. High readthrough transcription is also linked to increased nucleosome occupancy and high levels of H3K36me3. Moreover, forcing ARS readthrough transcription promotes these chromatin features. Finally, replication initiation defects induced by increased transcriptional readthrough are partially rescued in the absence of H3K36 methylation. Altogether, these observations indicate that natural noncoding transcription into ARSs influences replication initiation through chromatin regulation.

Introduction: Despite recommendations from the WHO, antenatal care (ANC) coverage remains low in many low-income and middle-income countries (LMICs). Community health workers (CHWs) can play an important role in expanding ANC coverage through pregnancy identification, provision of health education, screening for complications, delivery of therapeutic care and referral to higher levels of care. Background: This study aimed to evaluate the potential additive effects of measuring serum interleukin-1 beta (IL-1β) and interleukin-6 (IL-6) on enhancing the predictive value of baseline serum high-sensitivity C-reactive protein (hsCRP) levels for predicting 12-week antidepressant treatment responses in patients with depressive disorders.

Methods: Serum levels of hsCRP, IL-1β, and IL-6 were measured at baseline in 1086 outpatient participants diagnosed with depressive disorders. Participants initially received monotherapy with antidepressants for the first three weeks, followed by a naturalistic, stepwise pharmacotherapy regimen administered every three weeks up to 12 weeks.

Patients with Parkinson's disease (PD) in the moderate and severe stages can present several walk alterations. They can show slow movements and difficulty initiating, varying, or interrupting their gait; freezing; short steps; speed changes; shuffling; little arm swing; and festinating gait. The Movement Disorder Society-Unified Parkinson's Disease Rating Scale (MDS-UPDRS) has a good reputation for uniformly evaluating motor and non-motor aspects of PD.
