Partially Naked Fluoride in Solvate Ionic Liquids.

Truly naked fluoride exists only in the gas phase. Fluoride can be stabilized by a complexing agent and an organic cation, resulting in anhydrous or dehydrated fluoride which is "partially naked." This partially naked fluoride enables fluorination reactions at much lower temperatures than hydrated fluorides. Here we show a simple method for preparing fluoride-based solvate ionic liquids (SILs) by mixing 1-alkyl-3-methylimidazolium (1-ethyl-3-methylimidazolium or 1-butyl-3-methylimidazolium) bromide, silver fluoride (AgF), and EG (1:1:1 in molar ratio) in dry methanol. Removal of the methanol produced anhydrous SILs, [CCim]F·EG and [CCim]F·EG. This is the first SIL reported that comprises fluoride. H NMR and infrared spectroscopy reveal fluoride hydrogen bonds with EG OH groups and cation aromatic H atoms but not cation tail group protons. Fluorination reactions on benzyl bromide show that [CCim]F·EG has high reactivity with reasonable yield under mild conditions, confirming the fluoride ion is partially naked.

Mouse transmembrane protein 2 (mTMEM2) has been identified as a hyaluronidase, which has extracellularly G8 and GG domains and PbH1 repeats; however, our previously study showed that human TMEM2 (hTMEM2) is not a catalytic hyaluronidase due to the absence of the critical amino acid residues (His248/Ala303) in the GG domain. Naked mole-rats (NMRs) accumulate abundant high-molecular weight hyaluronan (HA) in their tissues, suggesting decreased HA degradation. Herein, we designed a chiral, axially-twisted molecular scaffold (ATMS) using pyridine-2,6-dicarboxamide (PDC) unit as pivot, chiral trans-cyclohexanediamine (CHDA) residues as linkers, and pyrene residues as fluorescent reporters. R,R-ATMS exclusively adopted M-helicity and produced differential response in UV-vis, fluorescence, and NMR upon addition of tartaric acid (TA) stereoisomers allowing naked-eye detection and enantiomeric content determination. Circular dichroism (CD) profile of R,R-ATMS underwent unique changes during titration with TA stereoisomers - while loss of CD signal at 345 nm was observed with equimolar D-TA and meso-TA, inversion was seen with equimolar L-TA.
