A simple and rapid method was developed for the analysis of ofloxacin enantiomers by chiral ligand exchange-high performance liquid chromatography (CLE-HPLC). In addition, the influences of common cations (Ca, Mg, Fe, Zn) and the content of humic acid (HA) on the separation were investigated. The separation was carried out on a C column (25 cm×0.46 cm, 5 μm). The mobile phase was 20% (v/v) methanol aqueous solution containing 4 mmol/L L-isoleucine (as ligand) and 3 mmol/L CuSO. The pH of the mobile phase was 4.5, and the flow rate was 1.0 mL/min. The column temperature was 40℃, and the detection wavelength was 293 nm. The ofloxacin enantiomers (ofloxacin and levofloxacin) were separated within 18 min and the resolution () was 2.70. The results showed that metal cations and HA had no significant influence on the resolution of ofloxacin enantiomers. However, the measured peak area of ofloxacin enantiomers decreased particularly in presence of Fe and on increasing HA content. The proposed method can rapidly and efficiently determine ofloxacin and its chiral isomers in surface water, the influences of Fe and HA content should be considered for verifying the applicability of the method.

The synthesis of optically active compounds requires determination of ee, er, and enantiomeric purity. The aim of the present paper is to review the synthesis of several chiral derivatizing reagents (CDRs) in a rational manner, which were successful for the separation and isolation of enantiomers of a variety of active pharmaceutical ingredients and other important and useful racemates. Besides, the application of (i) certain enantiomerically pure amines, either directly or by incorporating each of them as chiral auxiliary in difluorodinitrobenzene or cyanuric chloride moieties to construct the CDR, (ii) (S)-ketoprofen and (S)-levofloxacin as chiral platforms, and (iii) a few isothiocyanates, have been suitably included.

A capillary electrochromatography (CEC) synergistic enantioseparation system based on a novel nanomaterial synthesized by chiral molecularly imprinted polymers (CMIPs) and chiral metal organic frameworks (CMOFs) was developed. Compared with CMIPs and CMOFs alone, the enantioseparation performance of ofloxacin (OFL) of the CEC with the novel nanomaterial as stationary phases was greatly improved. CMOFs with chiral recognition ability have synergize with CMIPs to greatly improve the chiral selectivity of the novel stationary phases in CEC.

The key to developing sensors for chiral drug determination is to exclude interference from enantiomers. In this study, metal-organic frameworks (MOFs) and molecularly imprinted polymer (MIP) were introduced to prepare a chiral sensor for levofloxacin detection. The MIP was electropolymerised on the surface of the Cu/Fe-benzene-1,3,5-tricarboxylate MOF (Cu/Fe-BTC)-modified Au electrode using levofloxacin as a template molecule.
