Isolation of polysaccharides from Dendrobium officinale leaves and anti-inflammatory activity in LPS-stimulated THP-1 cells.

Dendrobium officinale stem is rich in polysaccharides, which play a great role in the medicinal effects of this plant. However, little was known about the polysaccharides from Dendrobium officinale leaves. Two kinds of polysaccharides in the leaves, DLP-1 and DLP-2, were obtained by hot water extraction, alcohol sedimentation and chromatographic separation (DEAE-52 cellulose column and Sephadex G-100 column). The average molecular weights were determined as 28,342 Da and 41,143 Da, respectively. Monosaccharide compositions were analyzed using gas chromatography-mass spectrometer. DLP-1 was composed of D-(+)-galactose, DL-arabinose, and L-(+)-rhamnose with a molar ratio of 3.21:1.11:0.23, and traces of D-xylose, D-glucose, and D-(+)-mannose. DLP-2 was consisted of D-glucose and D-(+)-galactose with a molar ratio of 3.23:1.02, and traces of D-xylose, DL-arabinose. Then, we established inflammatory cell model by LPS acting THP-1 cells to investigate the anti-inflammatory effects of DLP-1 and DLP-2. The results indicated that DLP-1 (5 μg/mL) and DLP-2 (50 μg/mL) were effective in protecting THP-1 cells from LPS-stimulated cytotoxicity, as well as inhibiting reactive oxygen species formation. In addition, both DLP-1 (5 μg/mL) and DLP-2 (50 μg/mL) significantly suppressed toll-like receptor-4 (TLR-4), myeloid differentiation factor (MyD88) and tumour necrosis factor receptor-associated factor-6 (TRAF-6) mRNA and protein expression in LPS-stimulated THP-1 cells.

Objective: Senile pruritus is a specific type of itching that occurs in elderly persons. Previously, we assessed antagonism of the nonselective ligand-gated cation channel transient receptor potential vanilloid 1 (TRPV1; capsaicin receptor or vanilloid receptor 1) and attenuation of atopic dermatitis by the non-steroidal TRPV1 antagonist PAC-14028 in clinical studies. The findings led us to postulate that PAC-14028 may also reduce itching in elderly people by antagonizing the TRPV1 pathway.

The discovery of novel anti-leishmanial compounds is essential due to the limitations of current treatments and the lack of new drugs in development. In this study, we employed the Quasi Vivo 900 medium perfusion system (QV900, Kirkstall Ltd, UK) to simulate physiological fluid flow, allowing us to compare macrophage responses and therapeutic outcomes under dynamic versus static conditions. After 24 hours, phagocytosis and macropinocytosis decreased in all cell types under flow conditions compared to static cultures.

Background: Diabetic retinopathy (DR) is a prevalent microvascular complication of diabetes and a leading cause of vision loss among diabetic individuals. Retinal pigment epithelium (RPE) cells play a crucial role in the pathophysiology of DR by releasing cytokines and exosomal cargo, such as long non-coding RNAs (lncRNAs), that modulate local immune responses, maintain retinal immune homeostasis and influence macrophage polarisation. Recent studies suggest that lncRNA cancer susceptibility candidate 2 (CASC2) may be involved in the regulation of DR progression.

The ultrastructure of mitochondria is pivotal for their respiratory activity. Thus, the regulation of the assembly of the super-complexes (SCs) of the mitochondrial electron transport chain (ETC) might be a core aspect of macrophage immunometabolism during bacterial infection. In order to study the impact of infection by Legionella pneumophila on the configuration of mitochondrial complexes and SCs in human macrophages, we have adapted and combined different methods such as cell sorting of infected cells, magnetic isolation of highly pure and functional mitochondria, quality control of mitochondrial purity by flow cytometry, and BN-PAGE (Blue-Native Polyacrylamide Gel Electrophoresis) coupled to Western Blot using near-infrared (NIR) fluorescence.
