A novel Marinactinospora strain JX35-4 was isolated from red soil which was collected from Wushan, northern Jiangxi Province, China. Analysis of the 16S rRNA gene sequences showed that strain JX35-4 belongs to the genus Marinactinospora and formed a distinct phylogenetic clade with Marinactinospora thermotolerans SCSIO 00652 and Marinactinospora endophytica YIM 690053 with sequence similarity of 96.97% and 96.42%, respectively. The strain was Gram-positive and formed branched substrate hyphae with no fragmentation, and abundant aerial hyphae that differentiated into long spore chains, and short rod-shaped spores. Growth occurred at 20-45 °C, pH 7.0-12.0 and in the presence of 0-7.5% (w/v) NaCl. The genomic DNA G + C content was determined to be 68.3 mol%. The cell wall of strain JX35-4 contained meso-diaminopimelic acid and xylose. Polar lipids were diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, phosphatidylinositol mannosides and one unidentified phospholipid. The major fatty acids of strain JX35-4 consisted of anteiso-C and iso-C. Major menaquinones were MK-9(H), MK-12 and MK-10(H). Based on the polyphasic data, strain JX35-4 (= CGMCC 4.7382 = DSM 104977) is concluded to represent a novel species of the genus Marinactinospora, for which the name Marinactinospora rubrisoli sp. nov. is proposed.

Collective migration of cancer cells is often interpreted using concepts derived from the physics of active matter, but the experimental evidence is mostly restricted to observations made in vitro. Here, we study collective invasion of metastatic cancer cells injected into the mouse deep dermis using intravital multiphoton microscopy combined with a skin window technique and three-dimensional quantitative image analysis. We observe a multicellular but low-cohesive migration mode characterized by rotational patterns which self-organize into antiparallel persistent tracks with orientational nematic order.

Understanding the interplay between biology and mechanics in tissue architecture is challenging, particularly in terms of 3D tissue organization. Addressing this challenge requires a biological model enabling observations at multiple levels from cell to tissue, as well as theoretical and computational approaches enabling the generation of a synthetic model that is relevant to the biological model and allowing for investigation of the mechanical stresses experienced by the tissue. Using a monolayer human colon epithelium organoid as a biological model, freely available tools (Fiji, Cellpose, Napari, Morphonet, or Tyssue library), and the commercially available Abaqus FEM solver, we combined vertex and FEM approaches to generate a comprehensive viscoelastic finite element model of the human colon organoid and demonstrated its flexibility.
