The conformational analysis of spider dragline silks is difficult because of the amorphous character of the silks. In this article, the fractions of several conformations were determined for three 47-mer peptides, (Glu)(Ala)GlyGlyAlaGlyGlnGlyGlyTyrGlyGlyLeuGlySerGlnGlyAlaGly-ArgGlyGlyLeuGlyGlyGlnGlyAlaGly(Ala)(Glu), with three underlined C-labeled blocks using a C CP/MAS NMR method. The conformations of the C-labeled sites change significantly depending on the location of the labeled blocks when treated with trifluoroacetic acid, low pH, and freeze-drying. The conformations of Ala and Gly residues are strongly influenced by the specific conformation of the (Ala) sequence at the C-terminal side, but those of other residues, Ala and Gly, and Ala and Gly, are basically not influenced by the conformations of (Ala). Through hydration of the β-sheet peptide, sharp peaks with random coil could be observed depending on the position of the residue, and this result could be interpreted via the change in the Ramachandran map obtained from the molecular dynamics simulation.

Research Background: Authentic Mexican cheeses have potential health benefits, although there are few studies on their bioactive components. In this study, we analysed soluble extracts from añejo cheese from Zacazonapan (Mexico), obtained from two dairies (A and B) that used milk from cows of different breeds and differed in processing.

Although the function of several PG synthetases and hydrolases is well understood, the function, regulation, and mechanism of action of PG hydrolases characterised as lysostaphin-like endopeptidases have remained elusive. Many of these M23 family members can hydrolyse glycyl-glycine peptide bonds and show lytic activity against whose PG contains a pentaglycine bridge, but their exact substrate specificity and hydrolysed bonds are still vaguely determined.

Intrauterine hypoxia (gestation days 15-19, pO 65 mm Hg, duration 4 h) led to an increase in the expression of p53, beclin-1, endothelial NO synthase (eNOS), and caspase-3 proteins in cardiomyocytes and reduced the number of mast cells in the heart of 60-day-old albino rats. Administration of a non-opiate analogue of leu-enkephalin (NALE peptide: Phe-D-Ala-Gly-Phe-Leu-Arg, 100 μg/kg) on days 2-6 of the neonatal period decreased the severity of delayed posthypoxic myocardial reaction. The content of eNOS cardiomyocytes and the total number of mast cells of these animals did not differ from the control parameters; the content of p53 cardiomyocytes was significantly lower than in animals exposed to intrauterine hypoxia.
