AI Article Synopsis

  • ERG3 is part of the ETS family of transcription factors, which are known for their conserved DNA-binding capabilities, though the specifics of how they interact with DNA sequences remain unclear.
  • The study involved analyzing the structures of the ETSi domain of ERG3 and its DNA complex, revealing significant conformational changes in both, particularly at the autoinhibitory CID motif.
  • Comparison with previous structures indicates similarities and differences in the binding mechanisms of ETS class 1 proteins to promoter DNA sequences.

Article Abstract

ERG3 (ETS-related gene) is a member of the ETS (erythroblast transformation-specific) family of transcription factors, which contain a highly conserved DNA-binding domain. The ETS family of transcription factors differ in their binding to promoter DNA sequences, and the mechanism of their DNA-sequence discrimination is little known. In the current study, crystals of the ETSi domain (the ETS domain of ERG3 containing a CID motif) in space group P422 and of its complex with the E74 DNA sequence (DNA) in space group C222 were obtained and their structures were determined. Comparative structure analysis of the ETSi domain and its complex with DNA with previously determined structures of the ERGi domain (the ETS domain of ERG containing inhibitory motifs) in space group P622 and of the ERGi-DNA complex in space group P422 were performed. The ETSi domain is observed as a homodimer in solution as well as in the crystallographic asymmetric unit. Superposition of the structure of the ETSi domain on that of the ERGi domain showed a major conformational change at the C-terminal DNA-binding autoinhibitory (CID) motif, while minor changes are observed in the loop regions of the ETSi-domain structure. The ETSi-DNA complex in space group C222 forms a structure that is quite similar to that of the ERG-DNA complex in space group P422. Upon superposition of the complexes, major conformational changes are observed at the 5' and 3' ends of DNA, while the conformation of the core GGA nucleotides was quite conserved. Comparison of the ETSi-DNA structure with known structures of ETS class 1 protein-DNA complexes shows the similarities and differences in the promoter DNA binding and specificity of the class 1 ETS proteins.

In the search for new ultraviolet (UV) nonlinear optical (NLO) materials, two novel cadmium mixed halide compounds, (NH)CdClF and (NH)CdBrF, are successfully synthesized via hydrothermal methods. These compounds crystallize in the noncentrosymmetric (NCS) space group, R32, and are composed of distorted octahedral [CdXF] (X═Cl or Br) units, which extend into a 3D framework. Remarkably, both compounds demonstrate strong second-harmonic generation (SHG) efficiencies-3.

Spaceflight-induced multi-organ dysfunction affects the health of astronauts and the safety of in-orbit flight. However, the effect of microgravity on the kidney and the underlying mechanisms are unknown. In the current study, we used a hindlimb unweighting (HU) animal model to simulate microgravity and employed histological analysis, ischemia-reperfusion experiments, renal ultrasonography, bioinformatics analysis, isometric force measurement, and other molecular experimental settings to evaluate the effects of microgravity on the kidneys and the underlying mechanisms involved in this transition.

Probing London Dispersion in Proton-Bound Onium Ions: Are Alkyl-Alkyl Steric Interactions Reliably Modeled?

We report spectroscopic and spectrometric experiments that probe the London dispersion interaction between -butyl substituents in three series of covalently linked, protonated -pyridines in the gas phase. Molecular ions in the three test series, along with several reference molecules for control, were electrosprayed from solution into the gas phase and then probed by infrared multiphoton dissociation spectroscopy and trapped ion mobility spectrometry. The observed N-H stretching frequencies provided an experimental readout diagnostic of the ground-state geometry of each ion, which could be furthermore compared to a second, independent structural readout via the collision cross section.

The cadmium-rich intermetallic compounds RhCd ( = Ca, Sr, Y, La-Nd, Sm-Lu) were synthesized from the elements in sealed tantalum tubes. The elements were reacted in an induction furnace and the samples were post-annealed to increase phase purity and crystallinity. The RhCd phases crystallize with the cubic CeCrAl type structure, space group 3̄.

This paper sheds light on how spaces become contested sites for identity construction and negotiation to take place. Applying the Social Representations Approach, a qualitative study of 10 focus group discussions (n = 39), was conducted in Singapore, Malaysia and the UK to explore how, and why racialised identity construction changed in each socio-political context. The study challenged two underlying assumptions in social psychology: (1) that the meaning of the racialised category holds constant across time and space, and (2) there exists a pan-racial identification among Asian identities, for example, which at times allows for racialised categories to be manipulated as variables.
