Stepwise Synthesis of Quercetin Bisglycosides Using Engineered .

Synthesis of flavonoid glycoside is difficult due to diverse hydroxy groups in flavonoids and sugars. As such, enzymatic synthesis or biotransformation is an approach to solve this problem. In this report, we used stepwise biotransformation to synthesize two quercetin bisglycosides (quercetin 3--glucuronic acid 7--rhamnoside [Q-GR] and quercetin 3--arabinose-7--rhamnoside [Q-AR]) because quercetin -rhamnosides contain antiviral activity. Two sequential enzymatic reactions were required to synthesize these flavonoid glycosides. We first synthesized quercetin 3--glucuronic acid [Q-G], and quercetin 3--arabinose-[Q-A] from quercetin using harboring specific uridine diphopsphate glycosyltransferase (UGT) and genes for UDP-glucuronic acid and UDP-arabinose, respectively. With each quercetin 3--glycoside, rhamnosylation using harboring UGT and the gene for UDP-rhamnose was conducted. This approach resulted in the production of 44.8 mg/l Q-GR and 45.1 mg/l Q-AR. This stepwise synthesis could be applicable to synthesize various natural product derivatives in case that the final yield of product was low due to the multistep reaction in one cell or when sequential synthesis is necessary in order to reduce the synthesis of byproducts.

Basic leucine zipper (bZIP) transcription factors serve as crucial regulators in plants' response to abiotic stress; however, its function in grapevine heat tolerance is still largely unknown. Here, we undertook a comprehensive investigation of grape genome, leading to the identification of 65 VvbZIP genes, among which 16 VvbZIPs were significantly induced under heat stress. Overexpression of VvbZIP36 enhanced heat tolerance in grape calli, while virus-induced gene silencing (VIGS) of VvbZIP36 reflected thermal sensitivity.

This study aimed to develop patches containing quercetin-loaded microcapsules and to evaluate their in vitro and in vivo safety and efficacy in preclinical surveys. A set of in vitro experiments evidenced the virucidal activity of quercetin against the HSV-1-KOS (sensitive to acyclovir) and HSV-1-AR (resistant to acyclovir) strains, with improved outcomes upon the first. The patches presented a homogeneous aspect, were easily handled, had a suitable bioadhesion, and possessed mechanical properties of soft and weak material, besides a pH compatible with human skin.
