The current rapid development of novel therapeutic approaches in immune oncology (IO) and specifically in the field of immune checkpoint inhibition is accompanied by an equally dynamic development of novel biomarker approaches for the identification of responding/non-responding patients under IO treatment. In addition to the measurement of the expression of checkpoint ligands/receptors, complex molecular predictors are gaining increasing attention in certain IO treatment constellations. This includes the entity informed identification of molecularly defined biological tumor subtypes (e.g., microsatellite instable neoplasms), the measurement of tumor mutational load and immune cell effector signatures as relatively routine diagnostic compatible novel biomarker strategies. In addition, a multitude of even more complex molecular IO biomarker approaches is emerging. This development is accompanied by new patient selection strategies which are based on the simultaneous combinatorial evaluation of more than one parameter. This article provides a comprehensive overview on currently relevant aspects in the field of IO biomarkers.

Lewy body (LB) pathology is present as a co-pathology in approximately 50% of Alzheimer's disease (AD) dementia patients and may even represent the main neuropathologic substrate in a subset of patients with amnestic impairments. However, the degree to which LB pathology affects the neurodegenerative course and clinical phenotype in amnestic patients is not well understood. Recently developed α-synuclein seed amplification assays (αSyn-SAAs) provide a unique opportunity for further investigating the complex interplay between AD and LB pathology in shaping heterogeneous regional neurodegeneration patterns and clinical trajectories among amnestic patients.

Background: Lab users should display an understanding of microbiology tests and avoid inappropriate requests which may yield negative clinical consequences. Our research addressed diagnostic stewardship education and the perspectives of paediatric hospital doctors on their (1) self-perceived knowledge, (2) motivators for test ordering, (3) proposed educational interventions and (4) preferred teaching modalities.
Methods: An anonymous, mixed-methods, 34-item online questionnaire was distributed to paediatric doctors across three Irish hospital sites from March to May 2023.
