The synthesis of oxalate esters through a radical process, rather than the traditional ionic reaction, has been well developed in which the radicals induced by visible light are trapped by molecular oxygen and diazo compounds under room temperature. This reaction is operationally simple, mild, and shows broad substrate scopes in α-bromo ketones and diazo compounds.

Gold-catalyzed synthesis of decorated indoles has been developed through carbene insertion into N-H bonds of -allyl-2-(aryl/alkyl ethynyl)anilines using α-diazo compounds followed by cyclization and concomitant 1,3 migration of allyl fragments. The developed protocol tackles the inherited challenge of direct C3 functionalization and eliminates the need for a tertiary aniline precursor for the 1,3-migration reaction. The applicability of this transformation is showcased through the practical synthesis of analogs of small drug-like and pharmaceutically relevant molecules such as ibuprofen, estradiol, menthol, and borneol, The mechanism is well supported by control experiments and isolation of the reaction intermediate.

PPh3/Isocyanide and N2/Isocyanide Exchange: Pathways to Isolable Alkali Metal Keteniminyl Anions.

Keteniminyl anions hold significant promise for advancing ketenimine chemistry, yet their isolation has remained elusive until now. Drawing inspiration from recent advances in ligand exchange reactions at carbon, we report the successful synthesis of isolable alkali metal keteniminyl anions through substitution of the phosphine ligand in metalated ylides or the N2 ligand in a diazomethanides with isocyanides. The exchange reactions were found to proceed more rapidly with aryl isocyanides than with the more electron-rich alkyl-substituted derivatives and were also more efficient when starting from the diazo compounds.

The synthesis of bioactive oxazoles is often inefficient, and the reported methods remain largely unexplored for complex derivatives. To circumvent this issue, we have utilized a metal-free, photomediated [3 + 2] cycloaddition reaction between diazo compounds and nitriles, leading to a step- and reagent-economical synthesis of bioactive oxazoles. These exciting developments guided us in the efficient synthesis of oxazole-based natural products like annuloline, alkaloids like pimprinethine, labradorin 2, and oxazole-containing pharmaceuticals such as oxaprozin.

Advances in the Synthesis of Cyclopropylamines.

Chem Rev

March 2025

Institut de Chimie Organique et Analytique, ICOA, CNRS UMR 7311, University of Orléans, 45100 Orléans, France.

Cyclopropylamines are an important subclass of substituted cyclopropanes that combine the unique electronic and steric properties of cyclopropanes with the presence of a donor nitrogen atom. In addition to their presence in a diverse array of biologically active compounds, cyclopropylamines are utilized as important synthetic intermediates, particularly in ring-opening or cycloaddition reactions. Consequently, the synthesis of these compounds has constituted a significant research topic, as evidenced by the abundant published synthetic methods.

In this paper, we describe a new type of cross-coupling between simple diazo and vinyldiazo compounds that gives access to unusual allyldiazo products. Blue light discriminates two diazo compounds towards free carbene formation, triggering sequential cyclopropenation, (3+2) cycloaddition and ring opening rearrangement processes. This strategy involves an overall reshuffle of diazo functionality and olefinic carbons of vinyldiazo compounds with an extrusion of nitrogen.
