The Quest for MAP Kinase Substrates: Gaining Momentum.

Mitogen-activated protein kinase (MAPK) pathways are versatile signaling mechanisms in all eukaryotes. Their signaling outputs are defined by the protein substrates phosphorylated by MAPKs. An expanding list of substrates has been identified by high-throughput screens and targeted approaches in plants. The majority of these are phosphorylated by MPK3/6, and a few by MPK4, which are the best-characterized plant MAPKs, participating in the regulation of numerous biological processes. The identified substrates clearly represent the functional diversity of MAPKs: they are associated with pathogen defense, abiotic stress responses, ethylene signaling, and various developmental functions. Understanding their outputs is integral to unraveling the complex regulatory mechanisms of MAPK cascades. We review here methodological approaches and provide an overview of known MAPK substrates.

The recent emergence of bile salt hydrolase (BSH) enzyme as a therapeutic target reflects its unbound potential in mitigating hypercholesterolemia, obesity, and gastrointestinal issues. However, to bolster its industrial application, optimization of BSH assay lays the cornerstone for enhancing sensitivity, specificity, and reproducibility. The current study delved into optimizing the BSH assay parameters utilizing response surface methodology (RSM) and one-factor-at-a-time (OFAT) method for two novel, natural BSH producers, Heyndrickxia coagulans ATCC 7050 and Lactiplantibacillus plantarum ATCC 10012.

A novel Ru-FeO nanozyme with enhanced peroxidase-like (POD-like) activity was synthesized through a hydrothermal method. Ru-FeO nanozyme was effectively utilized for the detection of thiophanate-methyl (TM) using a colorimetric technique. The POD-like activity of Ru-FeO was found to be superior compared to FeO, Rh-FeO, and Pd-FeO.

Aripiprazole (ARI) is an atypical antipsychotic which is a substrate of P-glycoprotein (P-gp), a transmembrane glycoprotein that plays a crucial role in eliminating potentially harmful compounds from the organism. ARI once-monthly (AOM) is a long-acting injectable form which improves treatment compliance. D-Allose, a rare sugar, has gained significant attention not only as a low-calorie sweetener but also for its anticancer, antitumor, anti-inflammatory, antioxidant, and other pharmaceutical properties.
