Serologic testing using the indirect antiglobulin test (IAT) is known to be insufficient to determine the clinical significance or insignificance of a given antibody to red blood cells (RBC), particularly in cases of antibodies to high-prevalence antigens, such as anti-Ge or anti-Yt. An in vitro functional cellular assay, the monocyte monolayer assay (MMA), has been studied for more than 40 years for its potential use to differentiate between clinically significant and insignificant RBC antibodies. The MMA has recently been used to select donor blood for transfusion into patients having a serologically incompatible crossmatch, without any obvious sequalae. Thus, the MMA shows great potential for future use to select donor blood for transfusion in situations where antibodies to high-prevalence antigens or complex multiple alloantibody problems confront transfusion services. In this report, we review the history leading up to the current uses of the MMA, its optimization, and potential use for the selection of serologically incompatible donor blood for transfusion. We describe current ongoing work to document and improve the efficacy of the MMA. Finally, we briefly describe possible future directions to make the MMA more amenable to routine laboratories.

The Innate Lymphoid Cells (ILCs) are a family of innate immune cells composed by the Natural Killer (NK) cells and the helper ILCs (hILCs) (ILC1, ILC2, ILC3), both developing from a common ILC precursor (ILCP) derived from hematopoietic stem cells (HSCs). A correct ILC reconstitution is crucial, particularly in patients receiving HSC transplantation (HSCT), the only therapeutic option for many adult and pediatric high-risk hematological malignancies. Indeed, mainly thanks to their cytotoxic activity, NK cells have a strong Graft-versus-Leukemia (GvL) effect.

The immunomodulatory properties of hyaluronan and its derivatives are key to their use in medicine and tissue engineering. In this work we evaluated the capability of soluble tyramine-modified hyaluronan (THA) synthesized from hyaluronan of two molecular weights (low M = 280 kDa and high M = 1640 kDa) for polarization of THP-1 and peripheral blood mononuclear cells (PBMCs)-derived macrophages (MΦs). We demonstrate the polarization effects of the supplemented THA by flow cytometry and bead-based multiplex immunoassay for the THP-1 derived MΦs and by semi-automated image analysis from confocal microscopy, immunofluorescent staining utilizing CD68 and CD206 surface markers, RT-qPCR gene expression analysis, as well as using the enzyme-linked immunosorbent assay (ELISA) for PBMCs-derived MΦs.
