We describe a short total synthesis of ajoene, a major biologically active constituent of garlic. The instability of allicin as the only other known alternative starting material has led to the development of a reliable procedure for the synthesis of ajoene from simple building blocks that is also suitable for upscale operations.

Histone deacetylases (HDACs) are important epigenetic regulators of gene expression and various cellular processes, and are potential targets for anticancer therapy. In particular, HDAC8 is a promising therapeutic target for childhood neuroblastoma. Ajoene is an organosulfur compound found in crushed garlic that exerts its anti-cancer activity by S-thiolating cysteine residues on proteins. Its development is hampered due to limited bioavailability, so in this study, we synthesised analogues of ajoene to probe the significance of the ajoene vinyl disulfide/sulfoxide core with respect to cytotoxicity and blood stability. Polar side groups were also incorporated to improve aqueous solubility.

Rapid and more environment-friendly means of gold nanoparticle synthesis is necessary in many applications, as in ion detection. Leaf extracts have become effective and economical reducing agents for gold nanoparticle formation, however, effects of extract combinations have not been thoroughly investigated. With the exploitation of combined extract effects, gold nanoparticles were synthesized then functionalized and investigated to produce selected nanoparticle systems which are capable of detecting aqueous lead(ii) ions with minimum detection limits of 10-11 ppm.
