Elucidation of lid open and orientation of lipase activated in interfacial activation by amphiphilic environment.

Lipases have wide applications using as biocatalyst in numerous biotechnological and bioengineering fields, especially function at hydrophobic or amphiphilic interface. Previously, the lipase from Burkholderia ambifaria YCJ01 was significantly activated when immobilized on the amphiphilic environment. In this work, insights into the functional effect of amphiphilic surface on lipase activation are presented by molecular dynamic simulations. The notable open of "lid" (α5 region) and the displacement of "flap" region (α8 region) of the lipase are closely related with the activation mechanism of lipase, which makes the active site accessible. Strikingly, the hydrophobic analysis showed that most of the hydrophobic surface residues of lipase, as an interfacial enzyme, located at the "lid" and "flap" regions make the entry to active site naturally orient to the oil-water interface to achieve enzyme activation. Additionally, the analysis of Rg and hydrogen bonding interaction suggested that the amphiphilic environment benefits to the exposure of hydrophobic regions, especially the "lid" and "flap" regions, and the maintenance of the nonpolar environment of the active site. Observations from this work not only complement the activation mechanism of lipase induced by the amphiphilic environment, but also provide a reference for the engineering of immobilized media for interfacial enzyme.

Water-soluble π-conjugated luminescent bioprobes have been broadly used in biomedical research but are limited by the nonbiodegradability associated with their rigid C-C backbones. In the present work, we introduced three naphthalene monoimide (NMI)-functionalized amphiphilic fluorescent polyesters (P1, P2, and P3) prepared by transesterification of functional diols with an activated diester monomer of adipic acid. These polyesters featured a side-chain NMI fluorophore, imparting the required hydrophobicity for self-assembly in water and endowing the polymeric nanoassemblies with green fluorescence.

We developed antibiotic-based micelles with bone-targeting and charge-switchable properties (P-CASMs) for treating infectious osteomyelitis. The amphiphilic molecules are formed by combining ciprofloxacin (CIP) with ligand 1 through a mild salifying reaction, and spontaneously self-assemble into antibiotic-based micelles (ASMs) in aqueous solution. Acrylate groups on ligand 1 enable cross-linking of ASMs with pentaerythritol tetra(mercaptopropionate) via a click reaction, forming pH-sensitive cross-linked micelles (CASMs).

Alzheimer's disease (AD) pathogenesis is correlated with the membrane content of various lipid species, including cholesterol, whose interactions with amyloid precursor protein (APP) have been extensively explored. Amyloid-β peptides triggering AD are products of APP cleavage by secretases, which differ depending on the APP and secretase location relative to ordered or disordered membrane microdomains. The study involves the modification of a non-conventional starch isolated from the under-utilized variety of Chinese water chestnut (CWC (Eleocharis tuberosa) and integrating it to fabricate stabilized and curcumin-enriched Pickering emulsions with enhanced bioavailability, thermal stability, and retention of encapsulated curcumin. A time-efficient, semi-dried esterification method was used to prepare modified amphiphilic starches using 3, 6, or 9 % (w/v) octenyl succinic anhydride (OSA) and characterized through degree of substitution (DS), contact angle, particle size, scanning electron microscopy (SEM), X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and in-vitro digestibility. Moreover, Pickering emulsions were formulated using CWCS-OSA at 3 %, 6 %, or 9 % concentrations to serve as a carrier for curcumin to improve its water solubility and storage stability.
