N-methyladenosine (mA) and N,2'-O-dimethyladenosine (mAm) are abundant mRNA modifications that regulate transcript processing and translation. The role of both, here termed mA/m, in the stress response in the adult brain in vivo is currently unknown. Here, we provide a detailed analysis of the stress epitranscriptome using mA/m-seq, global and gene-specific mA/m measurements. We show that stress exposure and glucocorticoids region and time specifically alter mA/m and its regulatory network. We demonstrate that deletion of the methyltransferase Mettl3 or the demethylase Fto in adult neurons alters the mA/m epitranscriptome, increases fear memory, and changes the transcriptome response to fear and synaptic plasticity. Moreover, we report that regulation of mA/m is impaired in major depressive disorder patients following glucocorticoid stimulation. Our findings indicate that brain mA/m represents a novel layer of complexity in gene expression regulation after stress and that dysregulation of the mA/m response may contribute to the pathophysiology of stress-related psychiatric disorders.

Background: Hypotensive episodes detected by 24-hour ambulatory blood pressure (BP) monitoring capture daily cumulative hypotensive stress and could be clinically relevant to cognitive impairment, but this relationship remains unclear.
Methods: We included participants from the Systolic Blood Pressure Intervention Trial (receiving intensive or standard BP treatment) who had 24-hour ambulatory BP monitoring measured near the 27-month visit and subsequent biannual cognitive assessments. We evaluated the associations of hypotensive episodes (defined as systolic BP drops of ≥20 mm Hg between 2 consecutive measurements that reached <100 mm Hg) and hypotensive duration (cumulative time of systolic BP <100 mm Hg) with subsequent cognitive function using adjusted linear mixed models.

While polyethylene terephthalate glycol (PETG) is widely used in orthodontic appliances such as clear aligners and retainers, there is limited experimental data assessing its performance under functional stresses, such as those encountered during dental movements and palatal expansion. This study aims to evaluate the ability of PETG thermoplastic material to withstand deformation under functional and expansion forces, specifically within the context of orthodontic applications. To estimate the firmness of the screw within the appliance, a universal Instron testing machine was used to record the forces released by each activation of the expander within the upper part of 10 clear modified twin blocks (MTBs) made from PETG and compare it with that released by 10 conventional twin blocks (CTBs).
