The 6-kDa early secretory antigenic target referred to as ESAT-6 is a virulence factor secreted by Mycobacterium tuberculosis (MTB). This work describes a voltammetric aptasensor for ultrasensitive detection of ESAT-6. Reduced graphene oxide doped with metal-organic framework (MOF-rGO) was deposited on a glassy carbon electrode (GCE). This increases the immobilization of electroactive Toluidine Blue (TB) and facilitates the electron transfer from TB to the modified GCE. Platinum/gold core/shell (Pt@Au) nanoparticles were used to assemble thiolated ESAT-6 binding aptamer (EBA) on a modified electrode and to further amplify the response to TB. The modified GCE, typically operated at -0.36 V (vs. SCE), has a linear response in 1.0 × 10 to 2.0 × 10 ng⋅mL ESAT-6 concentration range, and the limit of detection (LOD) for ESAT-6 is as low as 3.3 × 10 ng⋅mL. It exhibits satisfactory specificity and reproducibility when analyzing spiked human serum. Graphical abstract Schematic presentation of a voltammetric aptasensor for Mycobacterium tuberculosis antigen ESAT-6 using a glassy carbon electrode modified with reduced graphene oxide (rGO) and a metal-organic framework (MOF). The limit of detection for ESAT-6 is as low as 3.3 × 10 ng/mL.

Objectives: Pediatric tuberculosis (TB) diagnosis is complicated by challenges in obtaining invasive respiratory specimens that frequently contain few Mycobacterium tuberculosis (Mtb) bacilli. We report the diagnostic performance of an Mtb antigen-derived peptide (MAP-TB) assay and its ability to monitor TB treatment response.
Methods: Study cohorts enrolled children who presented with presumptive TB at two hospitals in South Africa from 2012 to 2017 (157 children aged <13 years) and at community-based clinics in the Dominican Republic from 2019 to 2023 (101 children aged <18 years).

Tuberculosis (TB) is the second deadliest infectious disease worldwide. Current TB diagnostics utilize sputum samples, which are difficult to obtain, and sample processing is time-consuming and difficult. This study developed an integrated diagnostic platform for the rapid visual detection of Mycobacterium tuberculosis (Mtb) in breath samples at the point-of-care (POC), especially in resource-limited settings.

Background: Public health issues related to tuberculosis still exist. Because Xpert MTB/RIF Ultra is more effective than conventional TB diagnostic techniques are, it is now regarded as an emerging technology. The diagnostic accuracy of Xpert MTB/RIF Ultra for tuberculosis was assessed in this systematic study.

Bovine tuberculosis (bTB), a global zoonotic disease, causes negative effects on human and animal health. PhoP protein is a key regulator of pathogenic phenotypes in members of the Mycobacterium tuberculosis complex, which includes the causative agent of bTB. Despite extensive research on this protein focused in deciphering its regulatory role, little was explored about it as a diagnostic antigen.
