A novel bacteriocin-like protein and its structural gene (rap) were identified from Rhodococcus erythropolis JCM 2895. The rapA and B genes are located on a 5.4-kb circular plasmid, and were obtained using a modified suppression-subtractive hybridization method. The rapA and B genes were heterologously expressed in Rhodococcus sp. or Escherichia coli, and then characterized. The results indicated that RapA is a small, water-soluble, heat-stable antimicrobial protein, and that RapB is an immunity protein against RapA, estimated to be located on the cell membrane. RapA showed antimicrobial activity particularly against R. erythropolis, and the activity persisted even after SDS-PAGE analysis. For the heterologous expressed RapA protein, N-terminal amino acid sequence was also confirmed. This is the first report of a bacteriocin-like substance obtained from the genus Rhodococcus.

Growing interests in replacing conventional preservatives and antibiotics in food and pharmaceutical industries have driven the exploration of bacterial metabolites, especially those from strains with generally recognized as safe (GRAS) status, such as lactic acid bacteria (LAB). In this study, a supernatant cocktail derived from multiple LAB strains was prepared and its bioactivities-antimicrobial, antibiofilm, antioxidant, cytotoxicity, and stability-were thoroughly investigated. The cocktail's main components were identified using thermal and protease treatments, gas chromatography coupled to mass spectrometry (GC-MS), and flame ionization detection (GC-FID).

Antimicrobial peptides, such as bacteriocin, produced by probiotics have become a promising novel class of therapeutic agents for treating infectious diseases. Selected lactic acid bacteria (LAB) isolated from fermented foods with probiotic potential were evaluated for various tests, including exopolysaccharide production, antibiotic susceptibility, acid and bile tolerance, antibacterial activity, and cell adhesion and cytotoxicity to gastric cell lines. Six selected LAB strains maintained their high viability under gastrointestinal conditions, produced high exopolysaccharides, showed no or less cytotoxicity, and adhered successfully to gastric cells.
