In order to investigate the function of monkey 20α-hydroxysteroid dehydrogenase (20α-HSD), transgenic mice (tg) were produced, expressing enhanced green fluorescent protein (EGFP) under the control of the monkey 20α-HSD promoter. The expression levels and localization of EGFP and 20α-HSD were analyzed in immature testis and in placenta. In support of our recent publication, "Characterization of transgenic mice expressing EGFP under control of monkey 20α-hydroxysteroid dehydrogenase promoter" (Park et al., 2018) [1], it was important to characterize the function of EGFP and 20α-HSD in the ovarian luteal cells of tg mice. Here, the expression of EGFP and 20α-HSD in immature testis and placenta are presented. The expression level of EGFP and 20α-HSD were detected in the testes 1 week after birth, and increased dramatically at 8 weeks. Both of proteins strongly detected in the placenta on days 14, 16, and 18 of pregnancy. Immunohistochemical analysis revealed that EGFP was detected in the seminiferous epithelium and 20α-HSD was specifically localized in the seminiferous tubule at 8 weeks.

Previously, it was reported that mRNA/cationic liposome complexes (mRNA lipoplexes) composed of the cationic triacyl lipid, 11-((1,3-bis(dodecanoyloxy)-2-((dodecanoyloxy)methyl)propan-2-yl)amino)-,,- trimethyl-11-oxoundecan-1-aminium bromide (TC-1-12), with 1,2-dioleoyl-glycero-3-phosphoethanolamine and poly(ethylene glycol) cholesteryl ether, induce high protein expression in human cervical carcinoma HeLa cells. In the present study, the authors aimed to optimize mRNA transfection using TC-1-12-based mRNA lipoplexes. mRNA lipoplexes were prepared at various charge ratios (+:-) using modified ethanol injection (MEI) and thin-film hydration (TFH) methods and compared the protein expression efficiency after transfection of HeLa cells with the developed mRNA lipoplexes.

Dendritic and axonal plasticity, which mediates neurobiological recovery after a stroke, critically depends on the mitochondrial function of neurons. To investigate, in vivo, neuronal mitochondrial function at the stroke recovery stage, we employed Mito-tag mice combined with cerebral cortical infection of AAV9 produced from plasmids carrying Cre-recombinase controlled by two neuronal promoters, synapsin-I (SYN1) and calmodulin-kinase IIa to induce expression of a hemagglutinin (HA)-tagged enhanced green fluorescence protein (EGFP) that localizes to mitochondrial outer membranes of SYN1 positive (SYN) and CaMKIIa positive (CaMKIIa) neurons. These mice were then subjected to permanent middle cerebral artery occlusion (MCAO) and sacrificed 14 days post stroke.

Interleukin 6 (IL-6) is one of the cytokines found to be multifunctional and biologically effective, regulating immune and inflammatory response by interacting with receptors to transmit signals. In this study, the full-length cDNAs of IL-6 (named as NaIL-6) and its receptors IL-6R and gp130 (named as NaIL-6Rα and NaIL-6Rβ) of Nibea albiflora were acquired and they possessed the typical symbolic motifs similar to its teleost orthologues in multiple sequence comparisons. The phylogenetic trees showed that NaIL-6 and its receptors clustered with their counterparts in bony fish, and had the closest affinity to Larimichthys crocea.
