CRISPR-Cas systems constitute an adaptive immune system that provides acquired resistance against phages and plasmids in prokaryotes. Upon invasion of foreign nucleic acids, some cells integrate short fragments of foreign DNA as spacers into the CRISPR locus to memorize the invaders and acquire resistance in the subsequent round of infection. This immunization step called adaptation is the least understood part of the CRISPR-Cas immunity. We have focused here on the adaptation stage of DGCC7710 type I-E CRISPR4-Cas (St4) system. Cas1 and Cas2 proteins conserved in nearly all CRISPR-Cas systems are required for spacer acquisition. The St4 CRISPR-Cas system is unique because the Cas2 protein is fused to an additional DnaQ exonuclease domain. Here, we demonstrate that St4 Cas1 and Cas2-DnaQ form a multimeric complex, which is capable of integrating DNA duplexes with 3'-overhangs (protospacers) We further show that the DnaQ domain of Cas2 functions as a 3'-5'-exonuclease that processes 3'-overhangs of the protospacer to promote integration.

Article Synopsis
  • CRISPR-Cas systems are immune defenses in bacteria that protect against viruses and genetic elements; certain viruses, like ICP1, also have their own CRISPR-Cas systems to fight back.* -
  • The ICP1 virus's Cas8f protein is missing a crucial domain for DNA cleavage, which raised questions about how it manages to interfere with host defenses.* -
  • Research uncovered that Cas1, typically involved in adaptation, also plays a key role in DNA targeting during the interference stage by linking other proteins to the DNA, providing new insights into phage CRISPR mechanisms.*

Article Synopsis
  • - PaaX is a transcriptional repressor involved in the degradation of phenylacetic acid (PAA) in bacteria, and it gets activated when phenylacetyl-coenzyme A (PA-CoA) binds to it, releasing it from its promoter sequences.
  • - The crystal structure of PaaX from E. coli reveals a unique dimeric formation made up of three distinct domains, including a winged helix-turn-helix domain and a dimerization domain resembling the Cas2 protein.
  • - Analysis of PaaX showed that its stability and solubility are influenced by ionic strength, and under certain conditions, it forms a stable intermediate that retains some structural features while exposing hydrophobic areas; these

DnaQ mediates directional spacer acquisition in the CRISPR-Cas system by a time-dependent mechanism.

In the spacer acquisition stage of CRISPR-Cas immunity, spacer orientation and protospacer adjacent motif (PAM) removal are two prerequisites for functional spacer integration. Cas4 has been implicated in both processing the prespacer and determining the spacer orientation. In Cas4-lacking systems, host 3'-5' DnaQ family exonucleases were recently reported to play a Cas4-like role.

CRISPR-Cas adaptive immune systems uptake short "spacer" sequences from foreign DNA and incorporate them into the host genome to serve as templates for CRISPR RNAs that guide interference against future infections. Adaptation in CRISPR systems is mediated by Cas1-Cas2 complexes that catalyze integration of prespacer substrates into the CRISPR array. Many DNA targeting systems also require Cas4 endonucleases for functional spacer acquisition.
