It has been shown that tetrodotoxin inhibits GABA-receptor binding of muscimol with solubilized membranes of the rat brain in a competitive manner, with the inhibition constant amounting to about 3 nM. Muscimol has been also demonstrated to competitively inhibit 3H-tetrodotoxin binding with the same drug.

Initial proof-of-concept development of competitive lateral flow (LF) test strips involving red quantum dots (Qdots) is demonstrated with visible confirmation of the competitive displacement of Qdot-protein-saxitoxin (Stx) and -tetrodotoxin (Ttx) conjugates over 15 min. The measured limits of detection (LODs) for the best versions of the assays were in the 1-2 µg range when a bovine serum albumin (BSA)-Stx or -Ttx conjugate was used as the analyte for safety reasons and NIH ImageJ image analysis was applied. However, when one assumes that only one of the primary amines in BSA labeled with Stx or Ttx is needed as the "epitope" for capture and calculates the weight of the toxin adducts on an equal mole basis to that of 1 µg of toxin-BSA conjugate, the LODs are as low as 4.

Simultaneous detection of tetrodotoxin (TTX) and okadaic acid (OA) is important for seafood safety. In this work, a novel paper electrode-based near-infrared (NIR) light-responsive photoelectrochemical (PEC) immunosensor was constructed using AgS quantum dots (QDs) and NaYF: Yb, Er upconversion nanoparticles (UCNPs) matched with BiOI for the simultaneous detection of TTX and OA in aquatic products. A low-cost, easily prepared gold nanoparticle-functionalized paper-based screen-printed electrode with six channels was designed to immobilize OA and Ab of TTX.

The marine toxin tetrodotoxin (TTX) poses a great risk to public health safety due to its severe paralytic effects after ingestion. Seafood poisoning caused by the consumption of contaminated marine species like pufferfish due to its expansion to nonendemic areas has increased the need for fast and reliable detection of the toxin to effectively implement prevention strategies. Liquid chromatography-mass spectrometry is considered the most accurate method, although competitive immunoassays have also been reported.
