The human body defends itself from harmful agents such as microorganisms present in the environment, which otherwise damage the individual's health. This defensive mechanism is the immune response mediated by the activation of T-cell and B-cell antibody production. The body recognizes and destroys these antigenic harmful agents. The defensive response is altered sometimes in the presence of allergens, causing hypersensitive reactions. The allergens are antigens. The hapten-carrier adduct often behaves as an allergen-producing hypersensitivity with a manifestation of sign and symptoms those are at times lethal. is an artificial type of poison formed by combination of two nonpoisonous substances as depicted in the treaties of Ayurveda. It has identical characteristics to that of hapten-carrier adduct. In the present article, is substantiated with conceptual reference to hapten-carrier adduct and hypersensitivity.

Our understanding of IgE-mediated drug allergy relies on the hapten concept, which is well established in inducing adaptive reactions of the immune system to small molecules like drugs. The role of hapten-carrier adducts in re-challenge reactions leading to mast cell degranulation and anaphylaxis is unclear. Based on clinical observations, the speed of adduct formation, skin and in vitro tests to inert drug molecules, a different explanation of IgE-mediated reactions to drugs is proposed: These are (a) A natural role of reduced mast cell (MC) reactivity in developing IgE-mediated reactions to drugs.

Allergic drug reactions are currently a major public health problem affecting patient health and increasing healthcare costs. They are caused by interactions between a drug and the human immune system and result in symptoms ranging from urticaria or angioedema to those more serious such as anaphylaxis or anaphylactic shock. The most commonly accepted mechanism for immunological activation is based on the hapten hypothesis.

Background: Organic acid anhydrides (OAAs) are highly allergenic compounds used in the chemical industry. The OAAs probably act as haptens but the proteins that form conjugates with OAAs in vivo are still unknown. Conjugates between the anhydrides and serum albumins (SAs) have routinely been used when testing for OAA-specific antibodies.
