Genome-wide association studies (GWAS) help to identify disease-linked genetic variants, but pinpointing the most likely causal genes in GWAS loci remains challenging. Existing GWAS gene prioritization tools are powerful, but often use complex black box models trained on datasets containing unaddressed biases. Here we present CALDERA, a gene prioritization tool that achieves similar or better performance than state-of-the-art methods, but uses just 12 features and a simple logistic regression model with L1 regularization.

Article Synopsis
  • Bartter syndrome is a rare genetic disorder affecting kidney function by disrupting electrolyte reabsorption, leading to severe health issues if untreated, including fatal hyponatremia and hypokalemia.
  • The study focuses on Bartter syndrome type II, which is linked to mutations in the ROMK gene; researchers identified over 40 associated mutations but most remain poorly understood.
  • Utilizing genomic databases and computational tools, the study discovered four new mutations in ROMK that affect its stability and function, with some mutations leading to premature degradation of the protein while others demonstrated resistance to degradation.

Unlabelled: The human papillomavirus (HPV) is the most common diagnosed sexually transmitted infection in the world. The most frequent disease linked to HPV is cervical cancer as well as other cancers including those of the vulva, vagina, penis, anus, and oropharynx. Our research sought to evaluate the knowledge and attitudes concerning human papillomaviruses and their vaccine among students enrolled in Altınbaş University's faculties of health sciences.
