Sterols are essential eukaryotic lipids that are required for a variety of physiological roles. The diagenetic products of sterol lipids, sterane hydrocarbons, are preserved in ancient sedimentary rocks and are utilized as geological biomarkers, indicating the presence of both eukaryotes and oxic environments throughout Earth's history. However, a few bacterial species are also known to produce sterols, bringing into question the significance of bacterial sterol synthesis for our interpretation of sterane biomarkers. Recent studies suggest that bacterial sterol synthesis may be distinct from what is observed in eukaryotes. In particular, phylogenomic analyses of sterol-producing bacteria have failed to identify homologs of several key eukaryotic sterol synthesis enzymes, most notably those required for demethylation at the C-4 position. In this study, we identified two genes of previously unknown function in the aerobic methanotrophic γ-Proteobacterium that encode sterol demethylase proteins (Sdm). We show that a Rieske-type oxygenase (SdmA) and an NAD(P)-dependent reductase (SdmB) are responsible for converting 4,4-dimethylsterols to 4α-methylsterols. Identification of intermediate products synthesized during heterologous expression of SdmA-SdmB along with C-labeling studies support a sterol C-4 demethylation mechanism distinct from that of eukaryotes. SdmA-SdmB homologs were identified in several other sterol-producing bacterial genomes but not in any eukaryotic genomes, indicating that these proteins are unrelated to the eukaryotic C-4 sterol demethylase enzymes. These findings reveal a separate pathway for sterol synthesis exclusive to bacteria and show that demethylation of sterols evolved at least twice-once in bacteria and once in eukaryotes.

The Citri Reticulatae Pericarpium (CRP), is the aged peel of Citrus fruit, which contains phenols, flavonoids, and polysaccharides. This study aims to investigate dietary CRP supplementation on the growth performance, serum biochemical indices, meat quality, intestinal morphology, microbiota, and metabolite of yellow-feathered broilers. A total of 240 yellow-feathered broilers (1.

The polyene antimycotic amphotericin B (AmB) and its liposomal formulation AmBisome belong to the treatment options of invasive aspergillosis caused by . Increasing resistance to AmB in clinical isolates of species is a growing concern, but mechanisms of AmB resistance remain unclear. In this study, we conducted a proteomic analysis of exposed to sublethal concentrations of AmB and AmBisome.

Background And Aims: To explore the predictive value of the triglyceride-glucose(TyG)index combined with non-high-density lipoprotein cholesterol (Non-HDL-C) in coronary atherosclerotic heart disease (CHD).
Methods And Results: We retrospectively collected patients who were suspected of CHD and underwent coronary angiography in Yiwu Central Hospital and collected medical history, other serum biochemical evaluation and echocardiography from the enrolled population, Non-HDL-C and TyG indices were calculated, and their correlation with Gensini score was analyzed. Logistic regression analysis was used to analyze the risk factors of coronary heart disease, and ROC curves were plotted to assess the predictive value of CHD in subjects with single or multiple indices.

Olanzapine (OLZ) is widely used in the treatment of schizophrenia, and its metabolic side effects have garnered significant attention in recent years. Despite this, the specific side effects of OLZ and the underlying mechanisms remain inadequately understood. To address this gap, zebrafish (Danio rerio) were exposed to OLZ at concentrations of 35.

Evidence regarding the relationship between free triiodothyronine (FT3) and low-density lipoprotein cholesterol (LDL-C) remains limited. This study aimed to evaluate the association between FT3 and LDL-C levels in patients with type 2 diabetes mellitus (T2DM) who exhibit normal thyroid function. Between June 2022 and October 2023, a total of 3011 inpatients with T2DM and euthyroid status were continuously and non-selectively recruited from a Chinese hospital.
