During a study to identify zoonotic pathogens in northwestern Mexico, we detected the presence of a rickettsial agent in Dermacentor parumapertus ticks from black-tailed jackrabbits (Lepus californicus). Comparison of 4 gene sequences (gltA, htrA, ompA, and ompB) of this agent showed 99%-100% identity with sequences of Rickettsia parkeri.

Upon invasion into the host cell, a subset of bacterial pathogens resides exclusively in the cytosol. While previous research revealed how they reshape the plasma membrane during invasion, subvert the immune response, and hijack cytoskeletal dynamics to promote their motility, it was unclear if these pathogens also interacted with the organelles in this crowded intracellular space. Here, we examined if the obligate intracellular pathogen Rickettsia parkeri interacts with the endoplasmic reticulum (ER), a large and dynamic organelle spread throughout the cell.

In Brazil, spotted fever (SF) is caused by two species of Rickettsia, both of which are transmitted by Amblyomma ticks: Rickettsia rickettsii, which results in severe and often fatal cases, and Rickettsia parkeri, which causes a mild illness. This study focused on R. parkeri in Amblyomma ovale ticks from the Maciço de Baturité region, Ceará, Northeast Brazil, an area endemic for SF with mild symptoms.
