Imbalance of Peripheral Blood T Helper Type 17 Responses in Patients with Vitiligo.

There is growing evidence to suggest that Th cells play pivotal roles in a variety of chronic inflammatory diseases, including vitiligo. However, the exact role of different subsets of Th cells in the pathogenesis of vitiligo is still a question. The purpose of present study was to determine the mRNA expression level of Th17 master transcription factor retinoic acid receptor-related orphan receptors gamma (RORɣt) and cytokine mRNA and protein expression profiles of Th17 cells. 22 patients with vitiligo and 22 normal subjects were enrolled in the study. Gene expression profiles of freshly isolated peripheral blood mononuclear cells (PBMCs) were determined by quantitative real-time reverse transcriptase PCR (qRT-PCR). Plasma concentrations of IL-17A and IL-22 were also assayed using ELISA kits. The results showed that RORɣt, IL-17A and IL-22 mRNA expression were increased in patients remarkably compared to healthy controls (p<0.05). Furthermore, plasma IL-17A and IL-22 levels were also higher in vitiligo patients versus controls (p<0.001). These data suggest that a deregulated Th17 adaptive immune response may contribute to the pathogenesis of vitiligo.

Background: Transient receptor potential cation channel subfamily V member 2 (TRPV2) functions as a stretch-sensitive calcium channel, with overexpression in the sarcolemma of skeletal and cardiac myocytes leading to detrimental calcium influx and triggering muscle degeneration. In our previous pilot study, we showed that tranilast, a TRPV2 inhibitor, reduced brain natriuretic peptide levels in two patients with muscular dystrophy and advanced heart failure. Building on this, we performed a single-arm, open-label, multicenter study herein to evaluate the safety and efficacy of tranilast in the treatment of advanced heart failure in patients with muscular dystrophy.

Background: Microsatellite instability-high (MSI-H) metastatic colorectal cancer (CRC) patients are the dominant population in immune checkpoint blockade treatments, while more than half of them could not benefit from single-agent immunotherapy. Military training improves tibial density, structure, and estimated strength; however, men and women may adapt differently. Most work performed in military populations has assessed changes in bone health during initial entry programs, a timeframe at the beginning of a service member's career when bones may be more adaptable to a novel mechanical stimulus. The purpose of this investigation was to examine changes in tibial volumetric bone mineral density (vBMD), structure, and estimated strength, and biomarkers of bone metabolism (P1NP, osteocalcin, TRAP5b, sclerostin) between male and female candidates measured at the start and end of United States Marine Corps Officer Candidates School (OCS), a 10-week military training program attended by older service members (~ 25 y/o) who may have previous military experience.
