Vinblastine, a potent anticancer drug, is produced by (Madagascar periwinkle) in small quantities, and heterologous reconstitution of vinblastine biosynthesis could provide an additional source of this drug. However, the chemistry underlying vinblastine synthesis makes identification of the biosynthetic genes challenging. Here we identify the two missing enzymes necessary for vinblastine biosynthesis in this plant: an oxidase and a reductase that isomerize stemmadenine acetate into dihydroprecondylocarpine acetate, which is then deacetoxylated and cyclized to either catharanthine or tabersonine via two hydrolases characterized herein. The pathways show how plants create chemical diversity and also enable development of heterologous platforms for generation of stemmadenine-derived bioactive compounds.

Anderson-Fabry disease is a hereditary, progressive, multisystemic lysosomal storage disorder caused by a functional deficiency of the enzyme α-galactosidase A (α-GalA). This defect is due to mutations in the gene, located in the long arm of the X chromosome (Xq21-22). Functional deficiency of the α-GalA enzyme leads to reduced degradation and accumulation of its substrates, predominantly globotriaosylceramide (Gb3), which accumulate in the lysosomes of numerous cell types, giving rise to the symptomatology.

Objective: To investigate the effects of the exosomal miR-494 targeting phospholipinositol 3-kinase (PI3K)/protein kinase B (AKT)/rapamycin target protein (mTOR) pathway on proliferation, migration, and invasion of trophoblast cells.
Methods: Decidual macrophages were randomly divided into control group, mimic NC group, miR-494 mimic group, inhibitor NC group, and miR-494 inhibitor group. Each group was transfected with corresponding miR-494 mimic NC, miR-494 mimic, and inhibitor NC and miR-494 inhibitor, while the cells of control group were only replaced with fresh medium.

(1) Background: For the reconstruction of a human vagina, various surgical procedures are available that are often associated with complications due to their failure to mimic the physiology of the human vagina. We recently developed a vascularized, organ-specific matrix from healthy human vaginal wall tissue with suitable biomechanical properties. A superior graft would require further extensive colonization with autologous vaginal cells to reduce complications upon implantation.
