Background: CRISPR/Cas9 enables the targeting of genes in zygotes; however, efficient approaches to create loxP-flanked (floxed) alleles remain elusive.
Results: Here, we show that the electroporation of Cas9, two gRNAs, and long single-stranded DNA (lssDNA) into zygotes, termed CLICK (CRISPR with lssDNA inducing conditional knockout alleles), enables the quick generation of floxed alleles in mice and rats.
Conclusions: The high efficiency of CLICK provides homozygous knock-ins in oocytes carrying tissue-specific Cre, which allows the one-step generation of conditional knockouts in founder (F0) mice.

It remains a challenge for a simple and scalable method to fabricate ultrathin porous Janus membranes for stretchable on-skin electronics. Here, we propose a one-step droplet spreading phase separation strategy to prepare an ultrathin and easily collected Janus thermoplastic polyurethane (TPU) membrane within seconds. The metal-ion solvation structure mitigated migration kinetics to delay TPU solution demixing, promoting the further penetration of the coagulating solvent.

A novel analytical method was designed and developed that exhibited ultraviolet-visible (UV-Vis), fluorescence (FL), and resonance Rayleigh scattering (RRS) signals for straightforward and comprehensive determination of monoamine oxidase B (MAO-B) using polyethylenimine-functionalized silver nanoparticles (PEI-Ag NPs). Through a facile one-step experiment, and NaOH assisted, in an aqueous solution of 100 ℃ for 40 min PEI reacted with AgNO to generate PEI-Ag NPs with a yellow color and weak blue fluorescence. Interestingly, phenylacetaldehyde (PAA), a specific product of MAO-B, causes significant enhancement of the three optical signals of UV-Vis, FL, and RRS.

A label-free photoelectrochemical (PEC) sensor for detecting theophylline (TP) was exploited based on electrodes modified with a nanocomposite of polydopamine nanospheres (PDSs) and gold nanoparticles (AuNPs). PDS particles were prepared by oxidative autopolymerization, and their reducibility was utilized in one step to reduce the gold nanoparticles . HfO/ZrO-based ferroelectrics present tremendous potential for next-generation non-volatile memory due to their high scalability and compatibility with silicon technology. Unlike the continuous polar layers in perovskite ferroelectrics, HfO/ZrO-based ferroelectrics are composed of alternating polar layers with oxygen shifts and non-polar spacers, which leads to a distinct ferroelectric switching mechanism. However, directly observing the switching process has been a big challenge due to the polymorph feature of nanoscale fluorites and the difficulty in in situ imaging on light elements.
