Myofibrilar functional dysregulation in fish: A new biomarker of damage to pesticides.

Endosulfan (ES) modifies the ultrastructure of skeletal muscle fibers and causes changes to the swimming behavior of fish. The objectives of the present work were to evaluate, in fishes of Australoheros facetus, 1) the integrity of myofibrils (Mf) by the analysis of SDS-PAGE profiles, and 2) the functionality of Mf through the microscopically monitoring of the contraction and changes in Mg Ca- ATPase and Mg(EGTA) -ATPase activities. As expected, after the addition of the contraction buffer, control fish Mf contracted. On the contrary, Mf from fish exposed at 0.5 μg/L ES showed a partial contraction and none of the fish exposed at 10 μg/L ES contracted. As judged by its high Mg Ca ATPase activity and low Mg (EGTA) ATPase activity, control Mf showed good functionality. In Mf from fish exposed to 0.5 and 10 μg/L ES the activities of these enzymes were similar, suggesting denaturation or degradation of some component of tropomyosin-troponin complex. SDS-PAGE patterns of Mf from fish exposed to ES showed degradation of the myosin heavy chain and of tropomyosin. Similar values of lipid peroxidation (TBARS) were found in both control and exposed Mf, suggesting that lipid oxidation was not be related to the above-mentioned changes. The observed effects expand the knowledge of ES action in muscles and could be used as biomarkers of damage in fishes exposed to organochlorine compounds like the insecticide endosulfan.

Expanding toolkits of EPA/DHA enrichment from natural sources is essential for better satisfying increasing demands for them. Lipase K80, from Proteus vulgaris K80, showed an application potential in EPA/DHA enrichment, whereas no desired heterologous expression in generally regarded as safe (GRAS) hosts restricted its relevant applications. In this study, expression of lipase K80 in a well-reputed GRAS host, Pichia pastoris, was achieved and further enhanced via combining disruption of its C-terminal KKL motif with co-expression of N-Acetyltransferase Mpr1, with a cumulative increment of nearly 200 % in the secretion level and the volumetric activity.

Humans are exposed to toxic methylmercury mainly by consuming marine fish, in particular top predator species like billfishes or tunas. In seafood risk assessments, mercury is assumed to be mostly present as organic methylmercury in predatory fishes; yet high percentages of inorganic mercury were recently reported in marlins, suggesting markedly different methylmercury metabolism across species. We quantified total mercury and methylmercury concentrations in muscle of four billfish species from the Indian and the Pacific oceans to address this knowledge gap.

Migration routes and the depth patterns of anguillid eel larvae migrating long distances from spawning grounds in the ocean remain poorly understood. We used otolith stable isotope analysis to study the oceanic migrations of anguillid eels by reconstructing experienced water temperature histories of larvae. The otolith stable oxygen isotopes (δO) of recruited Anguilla japonica glass eels were analyzed to assess the relationship with the experienced water temperature of the early larval stage in laboratory experiments.
