Objective: α-Galactosidases are widely used in many fields. It is necessary to improve the production of enzymes through microbiological processes. The aim of this study was to construct recombinant Aspergillus niger strains with high α-galactosidase production.

Results: Two recombinant A. niger strains were constructed: AB and AGB. The recombinant AB strain contained the α-galactosidase aglB gene from A. niger with its native AglB signal peptide regulated by the glucoamylase promoter. In the AGB recombinant strain, the AglB signal peptide was replaced with the glucoamylase (GlaA) signal peptide. The extracellular maximum α-galactosidase activity of the AGB strain was 215.7 U/ml and that of the AB strain was 9.8 U/mL. The optimal conditions for α-galactosidase were pH 3.5 and 35 °C.

Conclusions: The GlaA signal peptide substantially increased the yield of secreted α-galactosidase in A. niger. This recombinant strain holds great potential for industrial applications.

Background: Blood-based biomarkers, especially P-tau217, have been gaining interest as diagnostic tools to measure Alzheimer disease (AD) pathology.

Methods: We developed a plasma P-tau217 chemiluminescent immunoassay using 4G10E2 and IBA493 as antibodies, a synthetic tau peptide as calibrator, and the Quanterix SP-X imager. Analytical validation performed in a College of American Pathologists-accredited CLIA laboratory involved multiple kit lots, operators, timepoints, and imagers.

Positively charged residues are commonly located near the cytoplasm-membrane interface, which is known as the positive-inside rule in membrane topology. The mechanism underlying the function of these charged residues remains poorly understood. Herein, we studied the function of cytoplasmic residues in corin, a type II transmembrane serine protease in cardiovascular biology.

Background: Glucagon-like peptide-1 (GLP-1) is a crucial incretin hormone secreted by intestinal endocrine L cells. Given its pivotal physiological role, researchers have developed GLP-1 receptor agonists (GLP-1 RAs) through structural modifications. These analogues display pharmacological effects similar to those of GLP-1 but with augmented stability and are regarded as an effective means of regulating blood glucose levels in clinical practice.

The EphA2 transmembrane receptor is a key regulator of cellular growth, differentiation, and motility, and its overexpression in various cancers positions it as a promising biomarker for clinical cancer management. EphA2 signaling is mediated through ligand-induced dimerization, which stabilizes its dimeric state via conformational changes in the extracellular region and is linked to the intracellular kinase region via the transmembrane (TM) domain. Similar to many receptor tyrosine kinases, the juxtamembrane (JM) region, located between the TM and catalytic domains, coordinates with the TM domain to facilitate signal transduction.

Glyphosate (Gly) is a widely used herbicide for weed control in agriculture, but it can also adversely affect crops by impairing growth, reducing yield, and disrupting nutrient uptake, while inducing toxicity. Therefore, adopting integrated eco-friendly approaches and understanding the mechanisms of glyphosate tolerance in plants is crucial, as these areas remain underexplored. This study provides proteome insights into Si-mediated improvement of Gly-toxicity tolerance in Brassica napus.
