A method for the separation and determination of lactose and sucrose using high performance liquid chromatography-refractive index (HPLC-RI) detection base on the prepared amino-bonded silica gel chromatographic stationary phase was established. The method was accorded to the content determination method in Pharmacopoeia of the People's Republic of China (2015). Then, a series of chromatographic behaviors of lactose and sucrose such as the retention times, resolutions and peak area stabilities were investigated on three different types of amino-bonded silica columns (300 mm×4.6 mm, 5 μm). The mobile phase was acetonitrile-water (70:30, v/v) with isocratic elution, the flow rate was 1.0 mL/min and the injection volume was 10 μL. The resolution of lactose and sucrose was 3.03 by isopropyl side chain protected amino column, and the peak shape of each target compound was good. The relative standard deviation (RSD) of peak area of lactose was 1.14%, less than 2.0% prescribed in the Pharmacopoeia of the People's Republic of China (2015). Therefore, this method meets the requirements of lactose content determination, and is suitable as the quality control method in the above mentioned Pharmacopoeia.

A series of amino-derived mixed-mode chromatographic stationary phases were synthesized based on porous mercaptopropyl-functionalized polysilsesquioxane mesoporous microspheres synthesized by a co-condensation of methyltrimethoxysilane (MTMS) and mercaptopropyltrimethoxysilane (MPTMS). Through controlling the ratio of MTMS and MPTMS, the modified stationary phases with different amino densities were prepared by a "thiol-ene" click chemistry reaction. The morphology, pore structure, and functional groups of the microspheres were characterized by scanning electron microscope (SEM), nitrogen adsorption-desorption test, Fourier transform infrared spectroscopy (FT-IR), elemental analysis, and zeta potential, respectively.

A method was established for the rapid determination of 28 prohibited and restricted veterinary drugs of five classes (sulfonamides, quinolones, chloramphenicols, nitrofurans metabolites and triphenylmethane) in fish and shrimp by ultra high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) coupled with simultaneous extraction and purification. After optimizing the sample pretreatment methods and chromatographic conditions, the samples were hydrolyzed with hydrochloric acid. The five kinds of residual drugs were simultaneously extracted with acetonitrile after nitrofuran metabolites derivatized with 2-nitrobenzaldehyde.

Quantitative structure - retention relationships analysis was applied to investigate the molecular retention mechanism of proteinogenic and non-proteinogenic amino acids on the amino acid- and peptide-silica stationary phases. Twelve stationary phases with chemically bonded amino acids of different types (glycine, alanine, phenylalanine, leucine, methionine, aspartic acid, and N-(9-Fluorenylmethoxycarbonyl)-O-tert-butyl-l-tyrosine) and chains lengths (amino acid, dipeptide, and tripeptide) were tested. In order to compare chromatographic properties of the prepared materials with the conventional columns, the amino-bonded phases (laboratory-prepared and commercial one) were also studied.
