Tissue distributions of Leu-2+ cells in the spleen and draining lymph-nodes in cases of clinical gastric cancer were investigated, with special reference to suppressor cell function. Significantly higher Concanavalin-A (Con-A) induced suppressor cell activities were evident in spleen cells (SCs), as compared with peripheral blood lymphocytes (PBLs). As for the tissue distribution, the proportion of Leu-2+ (cytotoxic/suppressor) cells within Leu-1+ cells was higher in the spleen than in the lymph-nodes without metastasis. On the other hand, in lymph-nodes with metastasis, the enhanced spontaneous suppressor cell activity was noted. In addition, the proportion of Leu-2+ cells within Leu-1+ cells was the greatest in the lymph-nodes with metastasis, among the lymphoid organs tested. In lymph-nodes without metastasis, lower suppressor cell activities were noted, and numerous Leu-3+ (helper/inducer) cells were present, while Leu-2+ cells were less frequent. NK cell activity against K-562 cells was enhanced by elimination of Leu-2+/OKT-8+ cells with complement-mediated lysis. These results suggest that Leu-2+ cells located in the spleen and lymph-nodes with metastasis may predominantly act as suppressor cells and interact with effector cells.

Branched-chain amino acids (BCAA) are essential for maintaining intestinal mucosal integrity. However, only a few studies have explored the role of BCAA in the modulation of intestinal inflammation. In this study, we investigated in vitro effects of BCAA on the inflammatory response induced by lipopolysaccharide (LPS) (1 µg/mL) in Caco-2 cells.

Lipidated peptides derived from intracellular loops 2 and 3 of the urotensin II receptor act as biased allosteric ligands.

Over the last decade, the urotensinergic system, composed of one G protein-coupled receptor and two endogenous ligands, has garnered significant attention as a promising new target for the treatment of various cardiovascular diseases. Indeed, this system is associated with various biomarkers of cardiovascular dysfunctions and is involved in changes in cardiac contractility, fibrosis, and hypertrophy contributing, like the angiotensinergic system, to the pathogenesis and progression of heart failure. Significant investment has been made toward the development of clinically relevant UT ligands for therapeutic intervention, but with little or no success to date.

Water-soluble complexes are desirable for the aqueous detoxification of cyanide. Molybdenum complexes with α-amino acid and disulfide ligands with the formula K[(L)MoO(μ-S)(S)] (L = leu (), met (), thr (), and ser ()) were synthesized in a reaction of [(DMF)MoO(μ-S)(S)] with deprotonated α-amino acids; leu, met, thr, and ser are the carboxylate anions of l-leucine, l-methionine, l-threonine, and l-serine, respectively. Potassium salts of α-amino acids (leu (), met (), thr (), and ser ()) were prepared as precursors for complexes -, respectively, by employing a nonaqueous synthesis route.

Two new cyclic octapeptides, mariannamides A (1) and B (2), have been isolated from Mariannaea elegans NBRC102301, a Pinus densiflora-derived filamentous fungus. Their structures were elucidated to be cyclo-(l-Leu-l-Pro-l-Pro-l-Leu-l-Ile-l-Pro-l-Val-l-Ile) and cyclo-(l-Leu-l-Pro-l-Pro-l-Leu-l-Ile-l-Pro-l-Val-l-Val) based on spectroscopic data and Marfey's method. Mariannamide A (1) promoted mRNA expression of sirtuin 1 (SIRT1) in C2C12 cells, a mouse skeletal muscle myoblast cell line, and showed the antimicrobial activity against Escherichia coli and Cryptococcus neoformans.

Fractionation and purification of the ethyl acetate extract of the culture of a sponge-derived actinomycete, species Call-36, resulted in the isolation and identification of a new diketopiperazine, actinozine A (), cyclo(2-OH-d-Pro-l-Leu) (), two new nucleosides, thymidine-3-mercaptocarbamic acid () and thymidine-3-thioamine (), together with cyclo(d-Pro-l-Phe) () and cyclo(l-Pro-l-Phe) (). The structure assignments of the compounds were carried out by interpretation of 1D and 2D NMR data and mass spectral determinations. The absolute configurations of and were determined by Marfey's method and by comparison of the experimental and TDDFT-calculated ECD spectra.
