Chloroplast transformation vectors require an expression cassette flanked by homologous plastid sequences to drive plastome recombination. The 16-23 plastome region was selected and using this region, a new species-specific plastid transformation vector CuIA was developed with pKSII as a backbone by inserting the 16- and -23 sequences from L. An independent expression cassette with gene encoding aminoglycoside 3'-adenylyltransferase with controlling elements is added into the - intergenic region that confers resistance to spectinomycin. An efficient plastid transformation in bitter melon ( L.) was achieved by bombardment of petiole segments. The frequency of transplastomic plants yielded using standardized biolistic parameters with CuIA vector was two per 15 bombarded plates, each containing 20 petiole explants. Integration of gene was verified by PCR analysis in transplastomes. Transplastomic technology developed may be a novel approach for high level expression of pharmaceutical traits.

Due to the endocrine toxicity, neurotoxic, and reproductive toxicity to organisms, the sources and risks of brominated organic pollutants have attracted widespread attention. However, knowledge gaps remain in the bromination processes of emerging phenolic pollutants in plants, which may increase the potential health risk associated with food exposure. Our study discovered that light induced generation and accumulation of more toxic brominated organic compounds (Br-org) in lettuce leaves under the stress of acetaminophen (ACE) than that without light, as evidenced by an increase in C-Br bond intensity in FTIR analysis.

The plastid-encoded RNA polymerase (PEP) plays an essential role in the transcription of the chloroplast genome. Here, we present a strategy to purify the transcriptionally active protein complex from transplastomic tobacco (Nicotiana tabacum) lines in which one of the PEP core subunits is fused to an epitope tag. We describe experimental procedures for designing transformation constructs for PEP purification, selection, and analysis of transplastomic tobacco plants.

Background: The St-genome-sharing taxa are highly complex group of the species with the St nuclear genome and monophyletic origin in maternal lineages within the Triticeae, which contains more than half of polyploid species that distributed in a wide range of ecological habitats. While high level of genetic heterogeneity in plastome DNA due to a reticulate evolutionary event has been considered to link with the richness of the St-genome-sharing taxa, the relationship between the dynamics of diversification and molecular evolution is lack of understanding.
Results: Here, integrating 106 previously and 12 newly sequenced plastomes representing almost all previously recognized genomic types and genus of the Triticeae, this study applies phylogenetic reconstruction methods in combination with lineage diversification analyses, estimate of sequence evolution, and gene expression to investigate the dynamics of diversification in the tribe.

SnRK1 (SNF1-related kinase 1), a member of the SNF1 protein kinase superfamily, has been demonstrated to play a role in plant growth and development, as well as in stress responses. In this experiment, the leaf senescence of 'Xintaimici' cucumber was simulated by dark treatment and studied using SnRK1 activator/inhibitor and transient transformation technology. The effects of SnRK1 on cucumber leaf senescence, reactive oxygen species (ROS) metabolism, chloroplast structure, and photosynthetic characteristics were studied.
