Activation of mast cells in skin abscess induced by Staphylococcus aureus (S. aureus) infection in mice.

The skin abscess is a common inflammatory disease that occurs following the ubiquitous S. aureus infection. In our study, a skin abscess murine model was established and the dynamics of mast cells chemotaxing was evaluated. In the S. aureus-infected mice, severe infiltration of inflammatory cells in the dermis were observed, and mast cells were markedly accumulated in the skins. Besides, tryptase, the marker for mast cells activation, has a positive correlation with mast cell activity. The mast cells identified in the tissues were likely to be activated since they were associated with cell degranulation and the presence of tryptase. Our results suggested that mast cells and its mediator tryptase contribute to the inflammation of skin abscess induced by S. aureus infection.

: Human β-defensin 2 (HBD2) is a protein that plays an important role in activating the immune system by modulating spinal pathways and the inflammatory response. According to previous research, HBD2 was proven to be important in chronic spontaneous urticaria (CSU) (their values were significantly elevated in CSU patients, with a significant correlation between HBD2 levels and the percentage of peripheral basophils, suggesting that elevated HBD2 levels may be a potential marker of basophil and mast cell activation), which led us to additional research on the HBD2 molecule in isolated chronic angioedema. The aim of this research is to examine HBD2 values in the saliva and serum of patients with isolated angioedema, as a potential biomarker of the disease.

Based on viral infectious diseases, innate immunity cells such as mast cells and basophils play a fundamental role in the pathogenesis of viral diseases. Understanding these mechanisms could be essential to better study practical therapeutic approaches not only to COVID-19 but also to other viral infections widely spread worldwide, such as influenza A, HIV, and dengue.
