: Fractalkine (FKN) is a cytokine which plays an important role in atherosclerosis and other inflammatory diseases. Studies have shown that FKN induces integrin-independent leukocyte adhesion to primary endothelial cells under physiological flow conditions. Further, increased expression of FKN has been demonstrated in atherosclerotic lesions induced by low shear stress. However, the signal transduction mechanisms involved in low shear stress-induced FKN upregulation are not well characterized. In this study, EA.hy926 cells were subjected to varying intensity of fluid shear stress for different time durations. Further, mRNA and protein expressions of FKN were assessed by quantitative real-time PCR and Western blotting, respectively. Upregulation of FKN expression, which was induced via activation of mitogen-activated protein kinases signaling pathway under conditions of low shear stress, was studied both in the presence and absence of inhibitors. Low shear stress (∼4.58 dyne/cm) for more than 1 h promoted FKN expression and activated the extracellular signal-regulated kinase (ERK)1/2, p38, and Jun N-terminal kinase (JNK) mitogen-activated protein kinases signaling pathways by their phosphorylation. Inhibitors of ERK1/2, p38, and JNK pathways downregulated the FKN expression. In this study, fluid shear stress affected FKN expression in endothelial cells via activation of ERK1/2, p38, and JNK in a time-dependent manner. Our findings serve to advance the theoretical basis for prevention and treatment of atherosclerosis.

Capsules, which are potentially-active fluid droplets enclosed in a thin elastic membrane, experience large deformations when placed in suspension. The induced fluid-structure interaction stresses can potentially lead to rupture of the capsule membrane. While numerous experimental studies have focused on the rheological behavior of capsules until rupture, there remains a gap in understanding the evolution of their mechanical properties and the underlying mechanisms of damage and breakup under flow.

In particular, the thixotropic properties of materials, which allow them to flow under applied external forces but resist flowing otherwise, have been reported to enable rapid and high-resolution printing owing to their self-standing and easily processable characteristics. Objective: The study aims to elucidate the mechanisms underlying plaque growth by analyzing the variations in hemodynamic parameters within the plaque region of patients' carotid arteries before and after the development of atherosclerotic lesions.
Methods: The study enrolls 25 patients with common carotid artery stenosis and 25 with tandem carotid artery stenosis. Based on pathological analysis, three-dimensional models of the actual blood vessels before and after the lesion are constructed for two patients within a two-year period.

Theories predicted that shear promotes desorption, but due to the presence of factors such as aggregation effects, it is difficult to observe how shear influences the adsorption and desorption of individual protein molecules. In this study, we employed high-throughput single-molecule tracking and molecular dynamics simulations to investigate how shear flow affects the adsorption kinetics of plasma proteins (including human serum albumin, immunoglobulin G, and fibrinogen) at solid-liquid interfaces. Over the studied shear rate range of 0 - 10 s, shear stress did not trigger the protein desorption.
