An efficient method for the isolation and purification of 12,13-dihydroxyeuparin from by high speed counter-current chromatography (HSCCC) was established in this paper. The ether extracts of were purified by HSCCC with a solvent system of hexyl hydride-ethyl acetate-methanol-water (1:2:1:2, v/v/v/v). The upper phase was used as the stationary phase and the lower phase as the mobile phase. About 8.4 mg of 12,13-dihydroxyeuparin was obtained from 200 mg of ether extracts from in one-step HSCCC separation, with the purity of 96.71%, as determined by HPLC. After methanol-water recrystallization, the purity of 12,13-dihydroxyeuparin reached 99.83%. Indano[60]fullerene bromide was isolated as a key intermediate in the synthesis of indano[60]fullerene ketone, an evaporable fullerene derivative. Based on the identified reaction mechanism, aqueous hydrobromic acid was used to achieve reproducible and high-yielding reactions. A scalable one-pot reaction from aryl[60]fullerenyl dimers to indano[60]fullerene ketone was also demonstrated.

Background: Cholesterol efflux capacity (CEC) of HDL (high-density lipoprotein) is inversely associated with incident cardiovascular events, independent of HDL cholesterol. Obesity is characterized by low HDL cholesterol and impaired HDL function, such as CEC. Bariatric surgery, including Roux-en-Y gastric bypass (RYGB) and sleeve gastrectomy (SG), broadly leads to improved cardiovascular outcomes, but impacts on risk factors differ by procedure, with greater improvements in weight loss, blood pressure, and glycemic control after RYGB, but greater improvements in HDL cholesterol and CEC levels after SG.

Background: Smooth muscle cells (SMCs) of the proximal thoracic aorta are derived from second heart field (SHF) and cardiac neural crest lineages. Recent studies, both in vitro and in vivo, have implied relevance of lineage-specific SMC functions in the pathophysiology of thoracic aortic diseases; however, whether 2 lineage-derived SMCs have any predisposed transcriptional differences in the control aorta remains unexplored.
Methods: Single-cell RNA sequencing and single-nucleus assay for transposase-accessible chromatin sequencing were performed on isolated cells from the aortic root and ascending aortas of 14-week-old SHF-traced () and cardiac neural crest-traced () male mice.
