Objectives: Thalassemia is the most common genetically inherited blood disorder arising from a defect in hemoglobin production, resulting in ineffective erythropoiesis and severe hemolytic anemia. While transfusion therapy corrects the anemia, it gives rise to secondary iron overload. Current iron chelation therapy performed using deferoxamine, and the efficiency of this drug was demonstrated here using the zebrafish animal model.

Methods: Zebrafish larvae were exposed for 3 days to iron [100 μmol L ferric ammonium citrate; 3-6 days post fertilization (dpf)]. Then, iron treated larvae were exposed to 100 μmol L deferoxamine for 3 days (6-9 dpf). Total tissue iron concentration in the whole larvae, assessed by three different assays; inductively coupled plasma mass spectrometry, colorimetry (spectrophotometry), and microscopy using iron staining followed by imaging and quantification.

Results: The three assays showed that iron treatment alone resulted in a significant increase in total iron. Deferoxamine treatment of the iron-loaded zebrafish larvae showed a significant decrease in total iron concentration.

Conclusion: This study presented a clear evidence of the effectiveness of zebrafish larvae to use as a tool to study iron overload and open the door for studying the efficiency of potential new iron chelating compounds other than commercially available ones.

The aim of this work is to provide a comprehensive set of biological tests to assess the biomedical potential of novel osteochondral scaffolds with methods proposed to comply with the 3Rs principle, focusing here on a biphasic Curdlan-based osteochondral scaffold as a promising model biomaterial. experiments include the evaluation of cytotoxicity, mutagenicity, and genotoxicity referring to ISO standards, the assessment of the viability and proliferation of human chondrocytes and osteoblasts, and the estimation of inflammation after direct contact of biomaterials with human macrophages. experiments include assessments of the response of the surrounding osteochondral tissue after incubation with the implanted biomaterial.

Mechanical forces continuously provide feedback to heart valve morphogenetic programs. In zebrafish, cardiac valve development relies on heart contraction and physical stimuli generated by the beating heart. Intracardiac hemodynamics, driven by blood flow, emerge as fundamental information shaping the development of the embryonic heart.

Medicinal plants are considered promising candidates for controlling parasitic pathogen in aquaculture. Our previous study demonstrated that the crude extracts of Pseudolarix amabilis exhibit promising anti-Dactylogyrus intermedius activity. However, the specific compounds responsible for the antiparasitic effects of these crude extracts remain elusive.

Mutations in ADGRV1 can cause seizures, but the mechanism remains unclear. The zebrafish model can be used to assess the functions of human ADGRV1 and its variant alleles during embryonic development. In this study, we summarized the phenotypic and genotypic characteristics of four children with ADGRV1 variation and based on this, we validated the ADGRV1 loss phenotype in an adgrv1-knockout zebrafish model.

Color vision, which is mediated by cone photoreceptors in vertebrates, is essential for perceiving the external environment. Bisphenol A (BPA) and its substitute bisphenol S (BPS) have been widely used worldwide, while the evaluation of their safety, especially the newly discovered visual toxicity mechanism caused by them in recent years, has not been clearly explored. In the present study, we investigated the effects of BPA treatment (1, 10, and 100 μg/L) on cone cell development and function to evaluate visual toxicity.
