A fast confirmation method was developed for the determination of the six markers of pericarpium papaveris, morphine, codeine, narcotine, papavarine, thebaine and protopine in foods, by TurboFlow online purification-ultra performance liquid chromatography-triple quadrupole/linear ion trap mass spectrometry (TF-UPLC-QTRAP MS). The sample was extracted with 0.10 mol/L HCl. After the procedure of removal of lipid with hexane, the extraction solution was analyzed by TF-UPLC-QTRAP MS. The main factors influencing the purification efficiency including TurboFlow column, mobile phase and elution solution were optimized. The six opium alkaloids were detected by positive electrospray ionization tandem mass spectrometry in the multiple reaction monitoring-information-dependent acquisition-enhanced product ion (MRM-IDA-EPI) mode, and quantified by solvent standard internal standard method. The limits of detection were 0.05-0.5 μg/kg and the limits of quantification were 0.2-2 μg/kg for the six opium alkaloids. The recoveries were in the range of 81.1%-98.6% with the relative standard deviations ranging from 2.9% to 15.7% (=6). The method is sensitive and accurate, and has been successfully applied to the detection of pericarpium papaveris illegally added in foods.

Ethnopharmacological Relevance: Papaveris Pericarpium, which is the dried husk of Papaver somniferum L., has been used as a phytomedicine to relieve cough, diarrhea and pain. The alkaloid codeine contained therein via biotransformation converts to morphine and potentially produces addictive and toxic effects.

Overuse and abuse of morphine (MOP), one of the main components of pericarpium papaveris, have attracted increasing attention in the medical field owing to its pharmacological and toxicological activity. Herein, we proposed a new electrochemical nano-biosensor for MOP detection based on surface-confined building of Au@Pt-centered and multi-G-quadruplex/hemin wire-surrounded electroactive super-nanostructures. The center Au@Pt was flower-shaped and irregularly protruded, allowing substantial loading of multiple G-quadruplex wire/hemin complexes on its surface to accomplish the assembly of electroactive super-nanostructures.

Papaveris pericarpium, a natural source of morphine and codeine, is the principal active component in many antitussive traditional Chinese medicines. We herein report the first PK study of papaveris pericarpium in human plasma and urine following oral administration of single (15, 30, 60 mL) and multiple dose (15 mL) of Qiangli Pipa Syrup (MOR 0.1 mg/mL, COD 0.

A method for the separation and determination of five major opium alkaloids (narcotine, papaverine, thebaine, codeine, and morphine) in pericarpium papaveris by pressurized CEC (pCEC) with monolithic column has been developed. Under the optimum condition, linear calibration ranges of narcotine, papaverine, thebaine, codeine, and morphine were obtained as 2-85, 2-85, 5-75, 10-65, and 10-65 microg/mL, respectively. LODs of these analytes were 1.
