Bovine mastitis caused by Prototheca spp. can be a disease of high significance because of economic losses and the potential risk to public health. The aim of our study was to evaluate enzymatic activity of Prototheca zopfii. For this study, we used 15 P. zopfii strains previously isolated from cows with clinical and subclinical mastitis in Poland. We determined enzymatic profile of Prototheca species using the API ZYM system. Of the enzymatic activities detected during the study, acid phosphatase, leucine arylamidase, naphthol-as-bi-phosphohydrolase, esterase, lipase esterase, valine arylamidase, alkaline phosphatase, and lipase C14 were found in high percentage of strains.

Acetyl-CoA Synthetase Short Chain Family Member-1 (ACSS1) catalyzes the ligation of acetate and coenzyme A to generate acetyl-CoA in the mitochondria to produce ATP through the tricarboxylic acid (TCA) cycle. We recently generated an ACSS1-acetylation (Ac) mimic knock-in mouse, where lysine 635 was mutated to glutamine (K635Q), which structurally and biochemically mimics an acetylated lysine. ACSS1 enzymatic activity is regulated, at least in part, through the acetylation of lysine 635 in mice (lysine 642 in humans), a Sirtuin 3 deacetylation target.

The recently discovered lysine lactylation represents a critical post-translational modification with widespread implications in epigenetics and cancer biology. Initially identified on histones, lysine lactylation has been also described on non-histone proteins, playing a pivotal role in transcriptional activation, protein function, and cellular processes. Two major sources of the lactyl moiety have been currently distinguished: L-lactyl-CoA (precursor of the L-lactyl moiety) and S-D-lactylglutathione (precursor of the D-lactyl moiety), which enable enzymatic and non-enzymatic mechanisms of lysine lactylation, respectively.

In the present study, a Bacillus subtilis expression system was used to overexpress the gene of nitric oxide synthase (NOS), and the NOS was subsequently purified and added to fermented sausages to assess its colouration ability. The results indicated that NOS activity in the nos-recombinant strain was approximately 58-fold higher than that in the wild-type strain (P < 0.05).
