This paper describes a computational method to simulate the non-linear structural deformation of a polymeric aortic valve under physiological conditions. Arbitrary Lagrangian-Eulerian method is incorporated in the fluid-structure interaction simulation, and then validated by comparing the predicted kinematics of the valve's leaflets to in vitro measurements on a custom-made polymeric aortic valve. The predicted kinematics of the valve's leaflets was in good agreement with the experimental results with a maximum error of 15% in a single cardiac cycle. The fluid-structure interaction model presented in this study can simulate structural behaviour of a stented valve with flexible leaflets, providing insight into the haemodynamic performance of a polymeric aortic valve.

Based on clinical proteome analysis, we hypothesized that mitochondrial fusion and biogenesis in aortic smooth muscle cells (SMCs) are crucial for regulating the progression of NOTCH1-related aortopathy. Here we demonstrate that SMC-specific Notch1 knockout mice develop aortic pathology, including stiffening, dilation and focal dissection.

Aortic stenosis is a prevalent disease that is treated with either mechanical or bioprosthetic valve replacement devices. However, these implants can experience problems with either functionality in the case of mechanical valves or long-term durability in the case of bioprosthetic valves. To enhance next generation prosthetic valves, such as biomimetic polymeric valves, an improved understanding of the native aortic valve leaflet structure and mechanical response is required to provide much needed benchmarks for future device development.

Thoracoabdominal aortic aneurysms (TAAAs) are rare but serious conditions characterized by dilation of the aorta characterized by remodeling of the vessel wall, with changes in the elastin and collagen content. Individuals with Marfan syndrome have a genetic predisposition for elastic fiber fragmentation and elastin degradation and are prone to early aneurysm formation and progression. Our objective was to analyze the medial collagen characteristics through histological, polarized light microscopy, and electron microscopy methods across the thoracic and abdominal aorta in twenty-five patients undergoing open surgical repair, including nine with Marfan syndrome.
