Six new cyclopenta[g]indoles were isolated from a West Australian sponge, Trikentrion flabelliforme Hentschel, 1912, and their structures elucidated by integrated spectroscopic analysis. The compounds are analogues of previously described trikentrins, herbindoles, and trikentramides from related Axinellid sponges. The assignment of absolute configuration of the new compounds was carried out largely by comparative analysis of specific rotation, calculated and measured ECD, and exploiting van't Hoff's principle of optical superposition. Five of the new compounds were chemically interconverted to establish their stereochemical relationships, leading to a simple chiroptical mnemonic for assignment of the this family of chiral indoles. The first biosynthetic hypothesis is advanced to explain the origin of the trikentrin-herbinole family and proposes a pyrrole-carboxylic thioester-initiated polyketide synthase mechanism.

Late Eocene siliceous sponge fauna of southern Australia: reconstruction based on loose spicules record.

Zootaxa

February 2015

Instytut Paleobiologii PAN, ul. Twarda 51/55, 00-818 Warszawa, Poland.; Email:

An abundant and diversified assemblage of siliceous loose sponge spicules has been identified in the Late Eocene deposits cropping out along the southern coasts of Australia. Based on the comparison of the obtained spicules with those of living sponges, representatives of at least 43 species within 33 genera, 26 families, and 9 orders of "soft" Demospongiae and Homoscleromorpha have been identified in the assemblage. Within the studied sediments, the spicules representing demosponge orders Poecilosclerida, Hadromerida, and Astrophorida were the most diverse.

Chemical investigations of two specimens of Trikentrion flabelliforme collected from Australian waters have resulted in the identification of four new indole alkaloids, trikentramides A-D (9-12). The planar chemical structures for 9-12 were established following analysis of 1D/2D NMR and MS data. The relative configurations for 9-12 were determined following the comparison of (1)H NMR data with data previously reported for related natural products.
