Adenosine is an ancient extracellular signaling molecule that regulates various biological functions via activating four G-protein-coupled receptors, A, A, A, and A adenosine receptors. As such, several studies have highlighted a role for adenosine signaling in affecting the T cell development in the thymus. Recent studies indicate that adenosine is produced in the context of apoptotic thymocyte clearance. This review critically discusses the involvement of adenosine and its receptors in the complex interplay that exists between the developing thymocytes and the thymic macrophages which engulf the apoptotic cells. This crosstalk contributes to the effective and immunologically silent removal of apoptotic thymocytes, as well as affects the TCR-driven T-cell selection processes.

G-protein coupled receptors (GPCRs) are typically expressed on the cell surface where they mediate extracellular signals from hormones, neurotransmitters and growth factors, among others. However, growing evidence support the intracellular localization of GPCRs, including mitochondria. In the present work, we assessed the presence and functionality of adenosine receptors in mitochondria by combining techniques such as western blotting, radioligand binding, electron microscopy, enzymatic activities determination, oxygen consumption measurement and 3D morphological analysis of mitochondrial networks.

Progesterone (P4) secreted from the cumulus cells mediates nongenomic signalling in sperm cells and induces capacitation and acrosome reaction in the oviduct. The induction of these events in bull spermatozoa is not fully understood. Recently, we reported the presence of membrane nongenomic P4 receptors in bull spermatozoa and the involvement of P4-nongenomic signalling through the involvement of complex signalling pathways with cannabinoid receptors in the regulation of bull sperm capacitation and acrosome reaction.

Microvascular ischemia, especially in the heart and kidneys, is associated with inflammation and metabolic perturbation, resulting in cellular dysfunction and end-organ failure. Heightened production of adenosine from extracellular nucleotides released in response to inflammation results in protective effects, inclusive of adaptations to hypoxia, endothelial cell nitric oxide release with the regulation of vascular tone, and inhibition of platelet aggregation. Purinergic signaling is modulated by ectonucleoside triphosphate diphosphohydrolase-1 (NTPDase1)/CD39, which is the dominant factor dictating vascular metabolism of extracellular ATP to adenosine throughout the cardiovascular tissues.
